Lately I've been having issues with my grain jars and even my monotubs. Really not sure what the issue is ๐
I'm not exactly new at this and have several years of experience working with bulk substrates. I stopped the hobby for about a year and picked back up and have had nothing but mediocre results the last several runs I've done.
Im using Rye Seed - Always rinse it several times and soak for around 18 hours then boil till I see a couple burst or untill they're soft enough, then strain and set out to fully dry on the outside. Then I'll leave them sit out on a large peice of plastic and move around till the steam is gone and sit till the outside is dry enough - Followed by a pressure cook at 15 PSI for 100 mins.
After letting them cool down completely for the night ill set up my flow hood the next day and turn it on for an hour prior to inoculation. After removing the tin foil ill wape every surface and the jars down with 91% alcohol and spray lysol in the air.
In the picture of the jars they were all done on 10/06 - the jar on the left was from spores. The 2nd jar done from grain transfer (Argentina strain) and the two next to that to the right were done from grain transfer as well (Treasure coast).
They've all been broken up as needed depending on growth. Up untill a week or so ago they were maybe 40% completed. I Moved them to a warmer area that is 70 - 75 F consistently and broke them up. They were previously kept in an area that was 65 F - 70 F.
No matter what strain I use my jars are never as vigorous and nice looking as the results I used to get.
Several strains and 50 plus jars lately and terrible results of stalling or little fruits. ๐๐ฃ Anyone know where I could get a isolate from?
For the monotub I use the bucket method with coir and verm and allow to cool completely (Usually the next day) then squeeze to FC and mix my jars and flatten the surface, put the lid on and allow to and sit till fully colonized. Then I'll introduce fresh air and start the fruiting process. The picture I posted It wasnt fully colonized but it already started pinning so I introduced fruiting. The picture as also of the 2nd flush (1st flush was the same mediocre results - wimpy and small). I always fan 3-4 times a day and make sure they never dry out. Even the tubs before this using differnt strains and 100% colonized ill have little wimpy fruits.
Just really not sure where Im going wrong lately? All my spores are ordered from the same vendor I've used in previous years S.D and I'm using the same technique and sterile procedures but having bad results. Completely discouraged at this point and feel like giving up.

Are you cleaning up the spores before going to grain? Agar?

What do you mean cleaning up the spores before going to grain? The spores Im using are brand new. Purchased them over this past summer and kept in the refrigerator.
Never done agar before.. have not the slightest clue where to begin with that/acquire the materials needed. Even if I did do agar every syringe I try just comes out with small fruits or really skinny. I have terrible luck.

Spores are inherently dirty from being exposed to the air. Start with agar to make sure you get a clean culture. from there you can transfer as well to get an isolate. youre not going to find one from someone.

Ive never had problems in the past using syringes to jars. I dont really have any issues with mold. My jars just stall or i end up small/skinny fruits if it is successful.
Do you recommend any specific agar I should try? Or a good guide to follow / where to order it from?
By isolate you mean I should find the biggest mushroom possible (In this case the most normal size one - still little compared to what they should me) and cut the tissue and transfer to an agar dish?

You isolate mycelium by cutting and transferring from one agar plate to another. Once you end up with a healthy (contam free), robust isolated culture of mycelium you can use that to go to grains and test for fruiting. I'm working on it now too.
If you already have a flow hood there's no reason not to do agar work tbh. Read TEKs from TC's like these:

Thanks man! Ill look into them teks and try it. Did you order the agar online or were you able to find it locally at any big name stores?

Thats alot of information there. Just looked at c10s agar guide and it looks interesting and intimidating all at once ๐
I'm gonna give it a try and see If at the very least I can learn something more that might help me out.
