Man you are going to have a sweeeeeet setup! Only thing I can suggest is try the Bod's tek and try Pasty EZ dialed tek and see which one works better. I tried Bod's unmodified first and didn't work well for my conditions while Pasty EZ dialed worked like a charm. Just to see what will do well in your conditions!

And use a liner when you grow, I started doing it and it helps soooo much with side pins.

Also Munch's blenderless LI works well to if you have an agar plate you want to test quickly. Just dump the agar plate in some sterilzed water, shake the shit outta it, and pour into grains. I see you are already gonna be working on LC which is dope but if you want a quick test try that out.

Good luck my friend! Your setup has me jelly already haha.

Im also going to add some shelves for the tubs, jars and equipment etc, just a cheap one from Ikea like this :

I have no advice to give cuz you'll figure it out at some point. Except maybe start small but you already started big.
Anyway... what I wanted to add is just a clarification. There is no such thing as pan cyan tropicalis. Those are two different species. Pan cyan and pan tropicalis.

Oh. Here's one. You need to have about 10 petries for every strain/species you put on agar. I don't mean start them on 10 plates at once. But if you have just 50 plates I won't start more than 5.

You got some unnecessary equipment but knowing to start with agar is going to be in my opinion the biggest hurdle in the hobby. Its totally simple also intimidating but the biggest boon.

Good luck.

I think agar is the easiest part of the hobby. It's literally 1+2 = 3. So many other aspects are "I'd love to explain it to you but you'll just have to figure it out through trial and error. But that's just me.

Around $2500 - $3000 probably couldve got some stuff cheaper but i got this flow hood from China for $184!  Shipping was around $300 so came out to around 500 bucks for a new flow hood shipping included.

I wasnt expecting anything amazing to be honest, but it turned out great.
I think its the same or similar model to what Myers Mushrooms sells here:

Somebody asked me to link it so here it is.

https://m.dhgate.com/product/new-arrival-2020-ffu-1175-fan-filter-unit/582794516.html

Although it seems to have gone up in price a little since i ordered seen here:

Following this because I am sort of on the same path! Some of us are fortunate enough to have some scratch to throw around, while some of us are poor and need to do a little at a time. There is no shame in anyone's game, but I like to have ALL the necessary (and unnecessary) equipment too! haha Just make sure to log your activity and use science (which I have a hard time following haha).

A lot of glassware and lab equipment can be found dirt cheap from china. Not always best quality but for the price it has been well worth it. Media bottles and plates I grab from Amazon for quicker arrival but yeah stirrer, beaker, buchner funnels, vac pump and more for insanely low prices.

Did the FFU you ordered come with a prefilter on the back?

I very much want to ditch my clunky, overweight flowhood in the near future. I could really free up some room by getting rid of the thing. I even think I cold set up a space in my crawlspace for doing inoculations if I had a FFU. Finally free up the spare bedroom

Yes it did come with a filter on the back

Be very careful with that heated stirrer... I got a cheap one from China and had it set to 72*F (I made sure it was in Fahrenheit) and left it on for a day... came back to it and it had boiled my LC... just a heads up..

Unsure of the air intake on that flowhood, but she needs air to blow, I believe that's a ceiling, if not a wall unit.  I think, she needs to come off the wall a bit. Only going to blow available air than is sucked. Going to burn out fans pretty quick. Need to bring her 4-5" min. off back wall, if air intake is on the back.

Ok thanks I  will make sure to move the table forward a bit,im going to mount the unit to the table so it wont be able to fall.

The air flow is good, tested all speeds with a lighter and on all areas of the table at different distances  it was sufficient enough to blow the lighter or put it out,but i will test it with the plates i pour as soon as i do that. To check for contamination rates etc.

✌️😁

I made and poured my first batch of Agar two days ago now and left them infront of the flow hood unwrapped for an hour or more and left 3 fully open to test for contamination,so far none of the plates have any sign of contamination and look surprisingly clear  I will post a photo when i get home.

How long is it recommended to wait with plates after pouring to make sure they are all clean?

Thanks and Merry Christmas to you all !

Here are my first batch of petri dishes on the third day. No contamination so far. Will be pouring more and i think i need to alter my wrapping technique as the parafilm has cracked a couple of days after wrapping.

I prefer glad cling wrap. It's cheaper, get's the same job done, is easier to work with and doesn't have a moisture content.

Parafilm cracks if it has been left unsealed and dries out. It may crack for other reasons (like stretching's too tight) but I don't really know because I don't use it. Looks like you didn't stretch it tight though.

Thanks ill try that if the problem persists.

I poured more plates today and put swabs onto two of the plates that looked the cleanest.
I also made some LC (dextrose and water) to have ready and sterilized some water to have on hand if needed.

Here is the second batch i poured. These came out crystal clear and no condensation after following D3s Perfect Transfers and Agar Tek. !

I took five plates and left them with the lids off infront of the flowhood to check for contamination rate etc.

Because i was impatient and only waited 4 days after pouring to add the swabs to the agar so their could be potential contamination there but hopefully not.. They are all clean looking just some from the first batch have a bit of condensation.

I have a large dehydrator on the way which should hopefully be enough to begin with 😁

And I'm assuming you have plenty of airtight containers for bulk storage too, right?
Of course we don't discuss the other method of making that much fungus disappear here, but the way things are looking, you're gonna have way more than you can eat or share before they get old

From Aliexpress - Joanlab a Chinese lab equipment supplier.
It works better than expected although it needs about 10 minutes to fully heat up,but when hot sterilizes in about 4-5 seconds.

Wow life is so much easier now! Lmao never again will i parafilm 😂
I just cut a piece off the glad wrap roll to size and wrap the plates with that, way easier, looks cleaner and is much cheaper!

Damn OP, it took me almost two years to get all of the stuff you showed in your setup. Still haven't popped for the IR sterilizer, it's the last thing I want but don't have. All of this before so much as growing a single mushroom?!?!?

POWER!

We are kindred spirits it looks like. I like to go big right out of the gate too, I just had to start where I could and expand to bigger and better equipment as my money situation got better. My first grow was over 10 pounds dried and I started out using an SAB. If I hadn't lost 50 percent of my spawn when I first started out it would have been way more lol, it looks like you are on the right track to be the kingpin of mush in your area in no time. A non-profit kingpin of course.

All the homeless people, and veterans in my area know me as the guy that gives them free mush.

I'll be keeping an eye on you my friend, big things, and many contributions to the community seem to be in your future.

Yo bro, since you are in a position to equip yourself so thoroughly "like it ain't no thang", you should get an AA 75x when you can. You will LOVE it.

They are plug-in sterilizers and you can set them up with a timer. If you are planning on going relatively big it will change your life.

After my flowhood, it's the best thing I own. Might be close actually.

I definitely thought about getting that exact PC but at the end of the day the cost of it was too much at the time and the PC i currently have is sufficient for the time being but i will definitely be getting one some day in the future! 🍄🙏

One question if anybody can answer;

My PC has a release valve set at 24psi that automatically releases and holds at that pressure.
Is this too high pressure for PCing agar or grain?
(Ive currently been releasing pressure manually through the other release valve when around 17psi)

Thank you🍄

So ive decided to go with Corn instead of Rye as i have a few bags of the rye currently colonizing and im finding it difficult to see if they are clean or not due to the color of rye being brown and mold and contams tend to be green which makes it very difficult to see being colorblind,so im thinking corn will give me more transparency when looking for contams.

I will leave the discussion of rye vs corn for someone else, but if you are relying too much upon your eyes in this hobby you are going to be let down a fair bit. Your flowhood with proper sterile technique is the answer. Rather than choose a grain based on something like "easier to see contaminants on", I'd go with what you find to be the better performing grain in general, because even if the grain was see through, you aint gonna see all of the contaminants that can and will rear their ugly heads if you don't have sterile technique down.  Of the two you mentioned my money is on rye though, and I think that sentiment is pervasive, but popcorn does have it's fans. Do what works for you, but don't give too much credit to any particular grain when it comes to successful grows, or to being able to see stuff on it, or because of the contamination rate you may or may not experience, or because of anything else, because grain choice is a distant second to the end all be all of sterile technique. I may already be preaching to the choir to some extent as you've clearly done your homework broskie. You're on your way, I expect big things.

I dont expect to have to rely on it but again being colorblind makes it difficult to see contams on brown surfaces for me,actually almost impossible. atleast with corn i will have some fighting chance to see things as needed. I also have not been able to find rye in my local area so im definitely going to give corn a shot and if it turns out to be unfavourable i will give oats a try

thanks for the input buddy! Mush luv😎🍄

There is no grain when prepped and sterilized properly that will harbor contaminates. Corn works great. Oats work great. Rye, wheat, barley, tritacle, millet, milo, sorghum, spelt etc all work great.

Get a microscope for sure.

I haven't used a microscope since my schooling days and wouldn't know where to start. I want to get one as it has been discussed a fair bit recently, do you have any recommendations? What kind of specs should one look for on a microscope meant for this endeavor?

Yea definitely will be getting a microscope. I currently have a 400x zoom microscopic lense which works ok for taking photos but want a microscope asap for sure🍄

This is looking through the lid of a petri dish at 1600. Photos are taken with a 5mp microscope lens camera. I would recommend any good BINOCULAR microscope preferably with both bottom and top lights.

The field of view is about 10x that of the camera with the naked eye and real life resolution is clear as day.

This is an oil immersion view of zapotecorum? spores.

I see LOTS of mis-identification due to lack of microscopy capabilities. Very experienced cult members regularly call things mold and bacteria that are absolutely not either of those things... (picture 2 is Aspergillus Oryzae mold)

Great setup, great genetics, great thread. Very interested to see these Enigma cultures fruit.

Curious, does the chinese laminar flow hood come with a pre-filter?

That is the one thing I wish I had was a binocular scope. You are spot on about the top and bottom lights and a dimmer is useful too but man after a while, trying to hold one eye closed gets weird. I had the same problem with astrophotography. Those are muscles that you don't use like that often and they get tired and spastic when you force them to contract for long periods repeatedly.

With that said, a monocular scope is better than no scope and I still enjoy using mine.

Dimmer! very important. Good call.

My scope is binocular with 4 mag settings but it is only bottom lit and dimming. All my top lit viewing and photos are using a handheld flashlight next to the scope. It is really awkward with a flashlight.

The one I use was about 350 USD and came with a digital camera.

At least I don't have to melt, pour, and polish lenses like they did a couple hundred years ago...

I don't know where to file is so had to screen shot it from my phone's lock screen. This is the edge of a cut in the agar where I removed a wedge and the mic started to grow out from. It was taken with my cell phone held up to the scope so again visually it was much clearer.

Enigma LC turned out to be clean so that was added to 6 quart jars of sterilized corn to colonize, along with 4 agar plates in addition into two more quart jars of corn for a total of 8 quarts of Enigma on corn and 6 quarts of Enigma on rye.

APE has been transfered onto new plates for a hopefully nice and clean T2! 🍄

Everything is coming along nicely,Rye is almost fully colonized now and popcorn is well under way.

Ive put all varieties to Agar and will be isolating them and saving the best ones on slants for the future.

Hey im no expert but i might suggest not doing transfers with objects upwind. your petris are probably pretty clean but even so they create eddies in front of the petri your working with. also i like to work a little closer to the filter but maybe thats just my personal preference.

sorry if i missed it but what are the tools youre working with, i see a scalpel and ...?? an antler?? what is that? anyways nice setup, very clean.

Thanks for the tip! I will definitely take that into consideration, and keep that in mind for the future 🍄🤝

Its a custom made punch, made from deer antler, with inlays of mother of peal and turqoise lol 🤷‍♂️ Pretty extra i know but its extra long so i never have to worry about it heating up too much in the Bacti cinerator and looks great 🤌

I let the corn simmer for 45 minutes on high,rinse,strain and let dry for about 30 minutes,jar the corn,PC at 15-17psi for 1:45hrs - 2:00hrs. I do 1:50hrs🍄

Yeah I got a lot of hate when i first joined for using popcorn as well. Haven't used it in a while but I enjoyed working with it when I did. Easy prep and myc seemed unbothered by it. The large kernel size is the main reason I switched to WBS.

Enigma has been spawned.

The tub on the right is corn + coir (1:3)
The tub on the left is corn + rye + coir (2:3)

12qts coir 4qts corn
12qts coir 4qts corn + 4qts rye

Hi thanks for following and thank you for the kind words!

Lid on doesn't actually equal no FAE unless you are doing something special to seal up the tub. So it's unmodified?

The lid on, when sealed ,is air tight so that would mean there is no Fresh Air Exchange going on in the tub while colonizing .

So when i took that first picture it was still colonizing and hadnt been opened as i stated,so there was no fae.

I just realized that one of the bags i used to make a liner for my AA tub is a bio-degradable corn/plastic bag.

Will this matter? Thanks🍄

Awesome set up. I'm just here for the follow up! haha

Quite alot of growth in the last 4 days

The other tub of Enigma that was spawned on the same day has JUST started to fruit now 😂 not sure what the problem was there but it just didnt do anything for 1.5 month after fruiting. Oh well atleast its started now,i almost threw it out a couple times but something told me to wait. Its probably bacterial or something but well see how it progresses.

Averys Albino has been acting very strangely also, stalling after pinning completely and more pins just growing ontop of pins, not sure whats going on there but i guessed it might have dried out die to my rooms RH being a bit low,so i modified the tub,and will now wait to see, needs time to dial in tubs i guess. Good learning process.

Bro seriously, great job. I come back and check how you are doing while doing pretty much the same thing with a lesser budget myself.
I am a minimalist and seeing your lab design.. I love it.
I am curious; What are the ratios you use for grain spawn to the bulk substrate. I have 12 jars and 10x66qt monotubs, not sure how much of a grain spawn I should create with agar to make sure it's enough for the tubs.:mushroom2:

Thanks for following man! And good luck to you🍄

I used different ratios to test what was best, the tub that is going well had a ratio of 1:3.5 spawn - coir, and the other tub (that just started fruiting) had a ratio of 2:3 so around double the amount of spawn,it colonized very quickly but then didnt want to fruit for some reason and there was a single pin growing in the corner, and after a month it was still the same size so i plucked it and put it in the center of the tub just for lolz to see what would happen, also added a faux casing of coir and watered heavily. 2 weeks go by and now its pinning like crazy and the trasnfered pin that i plcuked from the side started growing again in the middle of the tub to my surprise!

Amazing things these shroomies.

True albino teachers

Rustywhyte

Put 1 month old KSAP Li to grain today, put a bit too much in the jars but looking  froward to see colonization times compared with agar.

And here is the contam Enigma tub,im pretty sure now that its just bacterial and not trich which is good.

Fingers crossed 🤞

That's going to turn green buddy. I would definitely isolate it or even salt it but really should probably toss it.

Well i honestly dont think so,but there are a few spots here and there that look suspect.  its growing really well at the moment though...as soon as i see anything start changing to green then ill toss it,if that happens.

What do you  think based on the below pics?

I dont have experience with enigma at all but those spots look totally like trich to me. That being said if it's been more than 24-48 hours and they're not green then they aren't trich, I don't think. From my experience it goes from white to green fairly quickly.

Maybe it's fluffy myc from need more FAE?

Harvested the first Enigma tub today, probably couldve gone a week longer but i felt it this morning and was a tiny bit soft here and there ,but hardly any difference really,it was still growing but i decided to snatch it while it was ripe😋

Very happy with the results - 1661 grams wet!

So ive put new genetics to agar today;

DC Mak (Melmak iso)
DC Mak cluster
Shakti (Albino malabar)

Also took transfers and harvested these chunky Kapes!

Now currently on agar and Li,awaiting grain we have;

TAT Clone
Pearly Gates multispore Liquid culture
DC Mak
Shakti
Kape Clone

In Tubs i am currently running ;

Enigma - ,one new tub that just finished colonizing and should start to fruit soon, also another tub that will be harvested soon (second flush).

APE - One tub, uncased - has started fruiting

Kape - One tub onto Fourth flush

Crooked Mystery(Tat iso from Dave wombat) - Starting to pin
