Liquid Culture (Help Please)

Today I'm attempting to make my first ever Liquid Culture and I have a few questions, If someone could help me or point me in the right direction I'd really appreciate it.



All I have is a large quart size jar and planned to make 400 - 500 MLS of LC - Should I add 1 teaspon of light corn syrup per 100 MLS and also use half CC of spores per 100 mls?



Example: 500 MLs - 5 tea spoons of light corn syrup and 2 and half cc of spores?

Would it be okay to boil the water and corn syrup or should I pressure cook it? Approximately how long should I do this for?

After inoculation how long does it take to see results? Anything special I need to do after its finished?

Sorry for so many questions.
Ive looked at a few different teks but still feel unsure about it. I'm trying to switch to LC because Ive had so many failed attempts lately with just spores.

1 teaspoon per 100 ml seems a bit rich, what is the sodium content of the corn syrup?

I use only like 2 grams of sitr for 300 ml you are doing about 4 grams per 100 ml that way.

I have always PC the LC but I could see it working with boiling the entire solution, (although less consistently) with the right method? (not sure.)

Results start happening in a day, you can begin to see the difference.



The darkest one on the right is Shiitake that are still incubating, the other 2 are done incubating and in a fridge at 40f. You can see how the solution clears from growth, but with clear syrup you wont get that effect.

Hi Teknix, thank you for the reply.

The sodium content is 30mg 1% per a serving size of 2 Tbsp (30ml) - Sugar 13g

The way I understood it from the teks I've looked at the math comes out to be 400ML x 0.04 = 16 grams of light corn syrup πŸ€·β€β™‚οΈ


Ill experiment with it and try different amounts per mixture.

I noticed alot of people use a stir bar or marble to break up the LC. Any suggestions on anything else that can be used or is it not necessary?

It could be that the sugar content is way less in corn syrup too, just seems like a lot.

I only put activated carbon rocks in it.

If you want the best chance at success you should PC it.

This Iberia Pure Cane turbinado sugar is 8 g sugars with 2 tsp, but 0 sodium :P

Should I pressure cook them at 15 PSI for 15, 20, 25 or 30 minutes? 

How will I know when the LC is 100% ready after inoculation? Should I just wait a couple weeks and stir it every few days?

Also how do you store your LC for long term use?

Again sorry for a zillion questions.. just want to get this right.

I PC'd these with some quart jars full of grains, it was about an hour, 30 mins at 15 psi would be fine for just LC. If you are firing up the PC, fill it up with things you need, especially tools you will use :smile:

Since you are growing from spores, it might take a couple days for you to see any change. 24 hours was for a culture. Spores straight into LC is kinda bad idea, you would be better off cloning a fruit and making an lc from culture.

You wan't proven genetics and cleanliness assured in an lc, not random assortments of monokaryotes and dikaryotes xD that might be contaminated. Sorry I missed that part before.

Multispore inoculation to a cake then to cloning and then making LC is where you are imo.

Unless you can make petri dishes and manually cross the monokaryotic spores, isolate dikaryotes and determine desirable growth characteristic from the dikaryotes you expand.

Thats way past my capabilities currently πŸ˜…. I wish I had stable genetics. I keep getting small fruits out of syringes Ive tried so far. Lately ive had several failed attempts with spores stalling.

You don't really want the monokaryotes colonizing the dikaryotes food tho. Monokaryotes don't fruit and they will compete with the dikaryotes that can.

If you clone a fruit properly, the mycelium is all dikaryotic.

Multispore into lc is kinda yolo move, but maybe you can learn something from it, or maybe even find some unforseen success, it is possible that you will get something worthwhile but the odds are definitely against it imo.

If all else fails it will be a learning experience 😁

Just took the two LC jars I just made out of the pressure cooker and they're super clear, gonna inoculate them tomarrow and see what happens.

Yep, you could also practice with live pure cultures from medicinal and gourmet mushrooms too! It's a big hobby so don't get stuck in the rut of growing cubes for monies. If you just invest a bit of money here and there into things you can use for years to come, you'll eventually have everything you need for this hobby. I still have an impulse sealer I got from a shroomerite when I was a noob! :lol:

Sounds like you're not ready for LC work then. LC is for people with skill dripping from their fingers. If you're not very successful at growing then LC is going to end up seriously pissing you off when it creates large scale failures.

Sugar LC sucks. Use something like potato and dextrose to make pda or malt extract to make mea.
