New to making transfer plates question
    #27004602 - 10/26/20 01:00 PM (3 years, 3 months ago)

I am having trouble keeping the agar that I am transferring "mycelium side up" which I have read is the preferred orientation. If the agar piece should end up on its side or even upside down is that a deal breaker?

I am using an agar punch to make 6mm plugs which are then transferred over to a new petri. I tried using an inoculating loop for transfer, but ended up using the tip of a scalpel.

Am I overthinking the process? Do you have a process that works for you?

It's not bad if it lands on the mycelium or on the side, I do think it makes the mycelium start growing abit weird but once it gets going it'll look normal.

Scalpals are the best for transfers in my opinion, once you've cut the square or wedge of agar try and slide the blade underneath the wedge so it's just sitting on the tip of your blade then place it down on the new plate kinda like how you picked it up from the original plate.

IME it  doesn't matter, so yeah don't overthink it. I've had pieces landing face down, to the side and they all grew perfectly!

If any of the said above is incorrect, please correct me! Still learning here and constructive criticism is the best

I try to get myc side down.  Sometimes I fumble it and it lands face up.  Then I swear and cuss.  But it never seems to really make a difference, haha.

It does not matter. If it lands on it's side, just use the scalpel to flip it this way or that. You could leave it on its side but that just increases the chances of it shaking free while wrapping and landing somewhere else in the plate.
