How to preserve mycellium on agar or in spawn jars?
    #27003940 - 10/26/20 01:47 AM (4 years, 2 months ago)

Hello,

I have two questions:

1. I'm wondering, how do I preserve mycellium so it will still be viable for the longest time? Both on agar and in spawn jars?

2. How long will it be viable for using that method?

Please note that I am talking both about cubes and gourmets (oysters and other colder-weather species mainly).

I have heard people before suggesting to make "slants" ... I don't understand why they would be any more effective at preserving mycellium that a normal agar dish, but anyway, okay. I don't have anything to make slants in, but sure.

Slants require less space and are generally more durable than typical disposable plates.

Hi,

I use agar plates to preserve my cultures.

Clean up your mycelium, put it on a few agar dishes, seal it up with parafilm, wait till agar has been colonised a bit,  put it into an airtight container (preferably with a desicant) and store it in your fridge.

I've stored my agar plates like that and they stay good for at least a year. I'm sure they'll be good for longer I just haven't tried that yet.

But actually slants are the way to go for long term storage.

Thanks!

Why do they need to be air-tight?

... and how long does just a normal spore print stay viable for? I would have though several years...?

RR states cold storage of colonized grain is one of the best methods. Better than slants except it does take up more room.

I use an airtight container in order to keep moisture out.

If you've got a print, that's even better. I think a member here grew mushrooms using a 15 year old print.

What's the point of this if spore-prints last even longer?

because you can store clones and other live cultures on slants for years without losing all your hard work. if you take a spore print of a clone, you're going back to random genetics.

When you have a proven culture, you can retain it and continuously( but not indefinitely)expand it.
A spore print has a multitude of random genetic material. Once a culture begins to senesce, multispore germination is where you go back to for new genetics to chose from.
