How can I prevent this PE blobing on these shoeboxes?  Should i harvest the blobs or leave them? is it too late to case? and If you recommend casing, I have some leftover hydrated coco from 3 weeks ago thats been in sealed cooler, can I use that or should i rehydrate fresh coco?  Thanks!

Anyone had any decent results cultivating PEU uncased?

With the gnat problem nearing control and bags being successful at warding off contamination for the time being I'm looking to expand out the 3 of these and do about 50-75 bags each. I know the PE will need casing but unsure about the PEU results without casing.

If my substrate is colonizing in a monotub, do I have to allow fresh air and how often and for how long?

Are you using modded or unmodded tubs? Introducing FAE as soon as you spawn seems to work for many people and I don't think the distinction of colonizing and fruiting conditions is really considered meaningful anymore. All my tubs right now are Pasty's EZ dialed monotubs, they work great--the holes provide passive FAE and have worked great for me so far. Give it a look.

Does anyone have a good cataloging and labeling system for agar? I'm going to start working with a few different varieties from MS and with GT clones and such and I feel like things are going to get out of hand quick lol. I'm already writing the date, inoculation method, variety, transfer, and numbering them consecutively but I feel like there's a better way.

Somewhat unrealistic/unnecessary but I'm trying to think of a way to label plates, grains and tubs in a way that lets me easily keep track of and trace the the final fruiting tub back to the plate that it originated from.

I guess it's not particularly difficult to just label the grains and tubs but I'm just wondering if anyone has a nice stream lined process of keeping track of everything that is consistent throughout each phase of the grow.

IIRC, stareatclouds had a good grain id tag that might help with something like this but I can't find it again.

Well I'll just throw this out for starters...

But there's ways I've gotten tripped up. Like in the moment of
loading shoeboxes with jars, I've suddenly realized I don't know
which jar in which tub. So now I have tape on the jar and the jar
tape label goes from jar to tub right then.

Dunno how realistic it is but I used to use red food dye for my agar, you could probably use like red, green, no food color, blue etc to be able to easily decipher varieties, just a thought...

Decimalise it.
I.e;
Ltc. 1.4.2.1 you read it backwards
So it's the 1st transfers from the 2nd transfer from the 4th transfer from the 1st transfer from germination.
Sorry that might be hard to read.
After that you can add notes like cl(a) clone (a)
Eventually you could go all the way around the Petri
LTC.1.4.2.cl(d).f2(a).2.1
In short is a t2 from the 2nd generation of a LTC multispore clone

Dude that is cool as hell, thank you for sharing. The color coordination is something I wouldn't have thought of, that's a great idea. Spreadsheet makes a whole lot of sense, I don't know why I was initially thinking of doing a text doc

How many plates/cultures are you usually working with? Do you find it reasonable keeping track of all those the way that you do now?

Also, great to see you're growing gourmets as well, I want to start some basic oysters on some straw I've had lying round but been so busy with the cubes I haven't gotten to it yet and I don't want to pay for a $25 syringe or LC when I could probably eventually find one to clone in the woods or even whole foods lol.

I was thinking of this too but I've heard some talk about the red and red3-containing food colors inhibiting myc growth so I'm kind of wary, not sure if there was a consensus reached on that though. I did have a whole bunch of magenta plates fail to germ spores but that could've been user error.

The problem with coloring is the myc eats it all pretty quickly.

For plates I just write on the lid, the name date and what it is. Like “tamp clone 1a, t2, 10-4.”

I just put a big whiteboard up to keep track of tubs I’m experimenting with.

Got another report to make.

Unmodded jars - no GE

Ive finally gotten around to make notes of my unmodded jars.
I've tracked the unmodded glasses for a time now.

My provocative claim:

You dont need any GE whatsoever, for 200 grams of grain in a glass jar to be colonized. You dont need a filter. They will finish in 2-3 weeks. Growth
can still look rhizo and good.

Some isolations will even colonize them in a time which i consider fast, which is surprising. But I dont have enough systematic experience to decide what is actually "fast" in cubensis colonization terms.

Is a month colonization for 500 grams of grain fast?

I will disclose that some jars seem to stall. But other jars, made from the same batch, but with different spawn, using the same jars, will finish in 2-3 weeks without any gas exchange.

Edit: the jars in question and colonization times mentioned are G2G transfers.

Edit 2: Ive tested that the jars in question are actually sealed.
when fully closed, I was unable to breathe air in or out of the unmodded jars/lids I use.

Edit 3: its of course possible that it affects yields. But the same jars colonize coir and fruit without problem.

Addendum: Its not my claim that GE doesnt improve colonization times, I know it does, and sometimes unmodded lids/jars stall. But I chalk that down to contamination, substrate and moisture, not to GE, because other jars in the same sitauation, will colonize fast and finish.

For people who grow and run into problems, if this "find" is true,
it has value. It allows you to cross out post inoculation as a contam vector.
For lazy, patient people, it also has value. It tells them that they can drop modding the lids, trading colonization time for not having to mod or make lids.

ACamb (Albino Cambodian) 5 is the germination plate. 5-1, 5-2, 5-3 are three separate transfers taken from the germ plate. 5-1-1 and 5-1-2 are two separate transfers from 5-1 plate, etc etc. FC = Fruit Clone, AC = Agar Pin Clone. I've neglected the shit out of my plate list and need to completely revamp it, but i keep hella notes. Then when it comes to grain jars/tubs, i only use one culture per variety at a time (only one APE or only one SAT, but run those side by side), and have a notebook to track results. On the jars/tubs, i just mark the variety and date, so it's difficult to mix them up and easy to track back to the plates. Just realized i did a bad job marking units under the shoeboxes, those numbers are Grams Wet/Grams Dry. Those were my very first shoeboxes ever, yes the yield was fucking garbage, shut up!

Yall really can't keep them in order if you just write the label on top? Variety and transfer number, and write sp if you want to spawn that plate when it's ready. That's a lot of work for something that you could just have on your plates.

Ha ha, and then what when the plates are gone?
Someone had a signature something like, "If you don't write it down, it's not science."

So anyway I'm just starting Munch's Shoebox Method
and as you can see I used an old SAB with two 4" holes and added four 2" holes (tomato paste can).

So, when will condensation show up?
I know the holes are too big to begin with, but when too start dialing in the FAE.
The shoeboxes have nice condensation on the walls, so I expect at some point the tub will.

  1. Perhaps I should just tape the shit outta it for starters and then dial back
when condensation shows up?

Finishing spawning 20 bags. Pretty damn enjoyable to do it this way it's like a shoebox with nothing to clean up afterwards.  1 quart WBS + 2 quarts CVG per bag. It'll be interesting to see the yields these are able to output and how long each bag will last before contaminating. Used to be I'd toss an entire mono if one corner went to shit so who knows what will happen if the same amount of material is spread out over four bags and doesn't need to all be chucked at once.

Still seeing the odd gnat or two in the house but not expecting them to be gone over night. Time will tell. I have two shoeboxes on my kitchen table which have nice fruit and no contamination and never saw a gnat in them so here is hoping.

Is it possible to get a clean clone tissue from a moldy fruit?

Found anomaly cube in my outdoor patch last night and wanted to see if it’s even worth trying to clone. It had this stringy green mold  growing from the ground to the cap like a spider web almost. Does mold grow into the fruit or is it more like bacteria where it just stays on the surface mostly?

I was planning on just rinsing it off, soaking it in a bleach solution for a couple minutes, and then rinsing again in 4 or 5 jars of sterilized water. Won’t waste my time with it if its already meshed though
