Quick and probably silly SOB question

I want to make a new still air box, my old one seems to work okay, but the arm holes are too close together and it's annoying.

For my new one, it seems to me it would be most comfortable and effective to actually have one arm hole in the side and one arm hole in the front, so your arms are actually coming together inside the box at right angles.

However, the thought occurs that this setup might serve to pump air through the box more than two holes in the same side.

Has anyone tried this and got any experience with a setup like that?

Hi.
My SAB has 2 armholes one each side of it, so i put each arm in each side of the box, kinda different of the one you are saying..
I also wonder if this brings any problems, like air flow of any kind..
But for now its working just fine, im still having contaminations in the agar sometimes but i guess its from the cultures itself, not from the flow of air.

But lets wait someone with more experience to say..

The two-side idea probably wont be very comfortable. I would recommend just getting a larger, longer clear plastic tub and place the arm holes on each side.

Also, u may have a point about the air pumping action if the arm holes are on different sides :strokebeard:

two holes in front will be better imo. if you have the holes on differnt faces, it will create an air draft which defeats the purpose of a SAB

I've had success making the smallest box possible, from cardboard covered with tape (to make it easier to wipe down). My box is big enough for only my left hand plus a few agar plates or 4oz jars.  I never put my right hand in the box, and I only glove my left hand. Instead I have a 3/4-inch hole that I can poke a tool through, like an inoculation loop or a syringe. I always hold and manipulate tools with my right hand, from outside the box. I use 4-inch-long syringe needles and my inoculation loop holder is about 10 inches long. So my tools always reach through the hole and into an agar plate easily even though my right hand is outside the box. The entire length of the tool I insert into the clean zone is flame-sterilized, every time it goes in. My left hand just moves plates around, wipes containers with alcohol and briefly lifts the lid to give access when needed.

I then extended this to a HEPA tunnel. A small HEPA filter flows clean air through the tunnel, which has the same geometry as the mini-SAB.

Benefits of this approach:

  • Less volume of air, therefore fewer spores afloat to start with.
  • Relatively larger surface area to help trap spores.
  • In the HEPA version, a small and cheap HEPA filter flows a lot of air through the tunnel.
  • A simple disposable glove covers almost the entire surface of the only hand that goes into the box.
  • Really easy to flame-sterilize the tools because the right hand isn't inside the clean zone. The alcohol lamp stays comfortably in open air, near the setup.
  • Really comfortable, because the right hand can work from many angles if the tool handles are long enough.  I can move my body around to give my left arm a comfortable angle into the one hole.


This approach isn't helpful for grain-to-grain or for dropping agar wedges into grain jars. But it's perfect for LC work. It makes LC work so easy, I'll never go back to doing grain-to-grain or agar-to-grain.

Ideally the SAB should be used with no air flow in the room anyway, so it won't matter where you have the arm holes as long as they aren't on top...

If the arm holes are big enough, and you're not making sweeping or fast movements, you're golden if you have the holes on the front, side or any combo. :smile:
