Watching the Myc Grow at 400x magnification 3

I am recording some T1 cultures on agar with a cheap 40/400x USB scope.
Here is a pic of 1 day old sample:

The video (placeholder):

On the laptop screen is a four-day-old sample of filthy's PESA at 400x. It was originally plated for germination via the ol' scratch and dump method, resulting in way too many germinated cultures on the plate.
I used d3monic's agar tek.
By tomorrow, I hope to have some good video of mycelium growing on this T1 plate.
I'll leave the camera recording overnight with the laptop plugged in... hopefully there will be no reason why the recoding gets cut short or lost... i was trying to record to an android phone earlier and ran into some software problems... only thing i could see getting screwed up here is a lack of HDD space due to this being a macbook air with less storage than a 12$ thumb drive.

my research suggests that photobooth is more likely to crash. video will be lost. looking for more "robust solution"

i appreciate the interest.
i am going to make an SAB to house this experiment... today, I'll try to capture a few hours of video- already have about an hour and a half on this last go but its takeing up space and will inevitably get jostled in the flow hood.
tomorrow, i'll set it up using a fully charged device in the SAB and try to get 24-48 hours of growth to time-lapse.

1)flame sterilize scalpel 2)scratch a small portion of a spore print off the foil 3)Dump/sprinkle loose spores onto agar plate
low tech. fast. always gets too many spores on the plate, making selection of transfer spots mental. My t1's have the amount of strains you'd hope for on germ plates... so it drags the isolation process out a bit longer than one might like.
caveat- this is my first time playing with mushrooms. Mycelium, anyway. Never seen a fresh cube in person.
Does anybody with experience know how constant direct light will affect the myc growth? now, they get 8 hours of light, unless i accidentally leave the hood light on overnight.

Longer than you like? So 50 transfers instead of 60? Isolates take a ton of work.
Streaking spores is best tho

that's why i used the specific language "isolation process"... not sure where in the process I am trying to get- new at this- but however many steps into the process, i get, further isolating and testing genetics, it will take longer when I start with messy germ plates.
my goal, more or less, is to arrive at some reliable and desirable cultures... fuck with tissue culture soon as I have fruit to work with... test out some different fruiting teks... create clean and reliable liquid cultures. But 50 transfers? No problem. I have 2-4 hours a day to work on agar projects at this point. And I like it. Suits me.
I've worked with prints every couple of days since the scratch and dump event with other prints and my technique is getting better and my sterile technique is getting into my muscle memory... I am going to try to do a serial dilution based on a tek from the guy from Fresh from the Farm Fungi on youtube.

i'm waiting on swabs. can you sterilize qtips? i have sterilization bags? or better in a jar?
i accidentally ordered 1000 disposable 10microliter loops... with syringes, i drop and then *try* to do the proper loop drag. have tried making a small amount of solution in a 30mm petri and then applying with a loop. so far, best plates from that method.
probably done germinating new strains, unless i receive other special genetics from generous OGs. I germed a PE swab from lovepotion yesterday along with some purple mystics commercially sourced spores.
i have my hands full with all these plates.

Just wrap em in foil(individually if motivated) and toss in a jar with the lid on. PC 15min.

I always throw a jar full of q tips wrapped up in foil in when I sterilize my agar. I usually cut them in half but sometimes I’m lazy and having the extra length is good 
I never liked using loops. I feel like swabs are better. But that’s just me

i wound up with less than 1 second of video from one hour of footage. the flow hood got bumped after about 1 hour.
its going to take 24 hours to get 20 seconds of growth.

Sweet thread idea. I've always swore i'd get right onto doing this. Direct light isn't really necessary but, if your goal is to speed up growth, it may help. I haven't done the tests needed to give you an exact answer on that. You can certainly tell that myc likes light though.

Not entirely undeserved. In my defense, the original camera required being hooked up to my laptop which i could not leave for longer that a few hours without using it for something else. I also had to take a part off to get it close enough to the plate and the adhesive that held the light ring inplace was oozing onto my plates. ALSO, the video kept dropping randomly. ALSO, without a solid mount, it moved around during recording. ALSO, the rez was shite. ALSO, the first new camera I tried was not much better.
The the NEW, new camera, which I spent $114 on is all self contained and solves every one of those problems.
I found a 32gb SD card in an old android phone in my car. Hope that's enough space for some overnight footage.
Next time I'll update with any issues that come up.

So here is a little teaser. Gonna take the entire weekend and a better sample to make anything super interesting.
I will use this one for the full weekend timelapse.

I use the scope periodically throughout the day so weekends are the longest i will ever get to record my, though I'd like to do a full week of growth. the camera will not record a whole plate... i al looking for a cam with the requires wide angle AND the resolution to do this job.

still having trouble getting video where the camera doesn't come out of focus over the course of the video. Also, media card was too small and a full weekend of vids kept getting overwritten every 6 hours...
but i am still figuring it out. In the meantime, my still game is getting better. Side lighting FTW. Multiple angles.

Ha. The struggle is real. I know the feeling. Those are sweet pics though. Learning to light petri shots and get them in focus is almost impossible for some folks.
