Azurescens->Agar->Grain * 1

Put a few drops of MS Syringe on PDA with charcoal.
This is about 3 weeks later. Please excuse the condensation, I know it's hard to see. Wasn't sure if I should take li off.




This appears to be mycelium, although I was expecting at least some contam. Should I just consider myself lucky, or is it possibly bacteria resembling mycelium? It is completely white and fuzzy. No rhizo but I heard it won't do that until it hits wood.

What should I do with it now? This is my first agar experiment, I was thinking just making more plates with it until im ready to noc up some wood jars..Theres no choosing sectors at this point, right?
will the three sepearate mycelium points be different in some way, or should i treat it as all the same?

none of my grain took from MS syringe and it's been 3 weeks. hoping for a revival but don't have hopes up. However, I would likw to make the best of this small amount of mycelium (if this in fact myc) in hopes of spawning a bulk bed come this winter/spring. So any help is appreciated!

Is this possibly very thick cobweb? Growth started a few days after introducing spores. The growth is completely white with no tinges any other color. I had an agar dish going from some old spores given to me and it was a light grey, clearly hairy and cobweb like a couple of days later. What’s growing in the Az dish is much different. Anyone have any input? Should I take the lid off for better pics?

I think you are doing good. Also for me Azurescens Mycelium is very thick and not rhizomorphic, very different to cubensis mycelium. Just make another transfer of the best looking parts and see what you get.

I'm currently germinating Psilocybe Azurescens from spores from a wild mushroom.
Got quite some contams, but after some isolation this is my current status:

Looks pretty similair to yours I'd say, just not as far along yet.
My next steps (also a first timer though) are innoculating some rye jars with the agar,
use this rye to bulk to pasturized wood chips, and use these woodchips to make a mushroombed outside. If you live in a temparate climate these mushies do best outside.
Not exactly sure though on what the best time is to start laying down your beds.
Maybe someone else knows?

Thanks man, looks great. I appreciate the affirmation! Our myc does look similar. And sounds like a good plan, this is what I was thinking as well. I live not too far from where they grow wild and I plan on growing it out until end of winter/spring then putting it in the ground and keeping moist through summer. Maybe planting a little grass on top and letting weeds do their thing to help retain moisture.
This is just based on what I read, but as of now it’s one big experiment! Good luck and thanks again for the pic

I'm soaking some rye now to transfer after sterilization!

Hi! It’s funny that you ask because I literally just noticed that the mycelium is now spreading to the grain. I’m using WBS (mostly millet) I also transferred a wedge of the colonized agar to a new plate just to keep things going. Here are some pics. You can see the myc barely starting to reach for the grain. Also, the agar in the grains now is from my first agar plate.. Everyone told me I must do at least 3 transfers to ensure a clean culture, I made one transfer to new agar, but instead of tossing the rest I thought I would just try to put it to grain to see what happens. Looking good so far. Maybe I just got lucky!

Looks good! mycelium seems to be colonizing quite well!

I ended up with this after a few transfers(guess it was like 5, but the spores were really dirty so kinda had to) :


The scrapings you see is me trying to put it on LC. The rest of the culture will be transferred to rye later today. Really hope to aquire a clean LC!

Let me know how it works out with the jars if you feel like it :smile: maybe we can help eachother along a bit every now and then!

What do you guys think? Seemed about 50% colonized with thick white mycelium, then some lighter, more hairy looking stuff started growing much faster in other parts of the jar. Thought it may have be cobweb but wasn’t sure.. shook jars about 3 days ago and this is what they look like now. Whatever this is grows very quickly and recolonized the entire thing within 72 hours. Is it just young, underdeveloped mycelium or some type of bacteria?





On an upside, look how beautifully this agar plate is coming along! If my jar is a bust, at least I have some of this to work with.. also have a bunch of stem butts ripping through cardboard now so no worries. Just curiosity. What a fun hobby:) Thanks y’all!

hmm hard to say from the picture, but that colonization time is a bit sus.
You could open one of the jars outside, to do a small smell test. I'm guessing (non contam) Azurescens also has this nice typical healthy mycelium smell to it.
Nice you keep the culture going anyway! Looks healthy and clean :smile:

I  tried making 2 LC's with 2% LME as carbohydrate source, but both still (after 10 days)  show no growth. Used the agar-biopsy method, i'm guessing something went wrong there.
Also tried making slants:

Made 4, 3 look clean.
Also put a bit of agar to rye, seems to be growing!


Also found some Azuresence hiking close to home and decided to try some clones from fresh mushrooms:

Not clean yet, but looking quite promising :smile: wonder if there's going to be a big difference in colonization speeds compared to the mycelium I got from the spores.

Small update;

Went from spores of last autumn to a second generation WBS spawn:


As most jars are pretty much done I decided to prepare the first woodchip sub today.
Here is a small pictorial + recipe. I'm not using any specifik tek (AFAIK), but have worked with wood for edibles before, so I think I can make some fair guesses.
Next to
1) Water
2) Gypsum
I'm using 3 types of wood substrates.
3) Beech sawdust
4) Small beech chips (<5mm)
5) Medium beech chips (<10mm)
My reasoning behind this is that I'm trying to create a somewhat PF tek like texture, where the bigger woodchips are covered with sawdust. This hopefully speeds up colonization (although I have no solid proof for this whatsoever).

The ingredients are combined in the following proportions:
1:2:3:4:5    -      10:1:12:10:8
This means that for every cup of gypsum, 12 of sawdust are used. And 10 of water (this doesn't cover all the substrates moisture, as you'll see).
First I throw the woodchips (4+5) together and soak them for an hour or two. In the meanwhile, the gypsum(2) and sawdust(3) are mixed with the before mentioned 10 parts of water(1) to bring it to field capacity. After two hours the chips are strained, and left to drain for about 30 mins. This should get them to field capacity. Breaking the biggest chips shows that everything is propperly moist and soaked.

Now dump it all together and mix well. Check if the moisture content is alright, and proceed to fill the filterbags.


The bags were sterilized for 120 mins., and are cooling down as I´m typing this post.
I´ll add the grainspawn (I´m thinking 500 ml/bag) in front of a flowhood tomorrow.
I'll update again once some worthy growth is starting to be visible! (or when all -so help me mushroomgods- fails)

I put some MS syringe down on PDA agar and was a bit worried that this was mold, but I'm thinking based on your guys pics, this looks normal for azurescens.  It just started growing a couple days ago, so not quite 2 weeks.

I would agree it is typical for early growth, but I have only been working with them since summer last year

That looks awesome!

Here's an update on my plates, I'm using a cloudy PDYA recipe, so the back lit photos aren't great. They are super dense and fluffy.  I just inoculated some oat yesterday, fingers crossed!
