These are some agar plates.
The following appears to have some yuck in it. Bacterial?



Is it normal for the mycelium to be fluffly? Also, the mycelium has kinda progressed in growth and grown over the possible contams, does that make it safe for transfer or are the contams still there intermingled with the myc?

Here are 2 other plates which imo seem healthy. What do you say?

The first three pics definitely have a shit ton of bacteria, everything else at least looks worthy of a transfer, but I’m still a noob so let’s see if anyone more experienced comes through to confirm. Even on the bacterial plates you could try to take transfers from the leading edge of the myc as far away as possible from the bacteria.

The general consensus is that the edges of plates are more likely to host hidden contams, so it’s not advised to let your myc grow all the way out and reach them before you transfer out. Seems like lots of people noc up jars and shit with their whole plate edges and all though, so take that as you will

Take the top off in a sab and tilt it towards the sab opening holes and get some better pics

Could be some super dense tomentose growth but I dunno. The lighter stuff definitely looks like contam.

Also, what kind of tape is that? Definitely not micropore considering I can literally see through the holes. I think you have the wrong kind of tape to be honest. If you can see through the hole, contams can get through the hole. I completely seal up my agar plates and their is always enough air for the mycelium to completely grow out.

Also never had an issue letting the mycelium grow to the edge of the dish. Lol, I'd like to see you stop it from growing to the edge. Just don't grab your sample from the edge or a previously contamed area and you should be good.

Its micropore but it melted during PC.  I suspect poor GE bc of that and hence fluffy myc, is that correct?

why do you have a hole in your agar dishes, no reputable tek ive seen so far calls for that

Its no pour agar it calls for a 6mm hole in the center and MP tape which melted during PC and also has a tiny hole in 1 of the plates

A strip of tyvek + micropore would be safer. Although i didnt have much contamination going the pasty plate 2 pieces of micropore tape.

Good catch - That's transpore tape, not micropore tape.  While both are 'surgical' tapes, they have completely different uses and are made from different materials.

From The manufacturer (3M),

"3M™ Micropore™Surgical Tape “Paper” TapeGentle, breathable tape with rayon backing. For general dressing applications and repeated taping on fragile, at-risk skin. Micropore tape— gentle, breathable"

"3M™ Transpore™ Surgical Tape “Plastic” Tape Transparent, perforated polyethylene film with bi-directional tear. Easily tears into very thin strips. For securing tubing and dressings that need to be monitored.Transpore tape—  clear, easy bi-directional tear"

Exactly, you want the paper looking stuff (it's actually more like a material). That's been the micropore tape I always use.

How do you guys know what contam you are dealing with? Like which kind of bacteria? Is this possible to even tell with the naked eye?

To be honest, it doesn't matter what kind it is. You don't want any.

Unless you want to study mold and bacteria, I wouldn't worry about the type.
