did my spore prints die? cant get spores to germinate
    #26977789 - 10/10/20 12:08 AM (3 years, 4 months ago)

hello fellow hobbyists. I have grown a few mono tubs of cubes successfully in the past.
I have two prints of my purple mystic cubes that are on tin foil and in zip lock bags sealed up. They are two years old, stored in a drawer in the house.

Recently I wanted to try to get these puppies going again. My agar agar recipe on petri dishes consists of the following.
5 grams of light dry malt
5 grams of Agar Agar powder
.5 grams yeast
250 ml water
I have followed BOD's Comprehensive Agar TEK every time I make my petri dishes and it has worked great with my recipe in the past.

SO I cant not get these spores to germinate on my agar dishes at all. It has been three weeks or longer. no mold or anything growing. I have tried three different times with both my spore prints on different dishes. I have waited about a week and a half each time. I use a sterile swab and roll it on the petri dish. I have even turned a couple of the dishes upside down I read that it could help. The dishes are kept at room temp, in the house.

Im not sure what to try next. These were some really fantastic fruits and would really like to get these ones going again.
what am I doing wrong?

refrigeration imo. I cold store all my prints and i've used prints about that age and they worked.

How much agar powder are you using? Softer (less agar powder) is better for germination.

When swabbing a print, I usually just press the swab to spores firmly, and the just kinda wiggle to get a dark mass of spores localized on an area of the swab, and streak with that. Then for the coup de gras, I jam the cotton tip into the agar. if that doesn't work you probably have to try and hydrate the spores in a syringe as your next step.

2years is not old at all, maybe they were improperly stored, got to hot or cold or whatever

I recently had a print done in a plastic petri dish, stored in a zip lock bag in the fridge, and after one month it was bone dry. The spores came off as flakes/needles. Put them on agar and they didn't germinate after 1 month. On the other hand I have prints in tinfoil stored just in my cupboard, which still germinate after 2 years. I think avoiding dehydration is essential, so you could also try to hydrate your spores first by soaking them in sterilized water or creating a spore syringe.
