I have been really conflicted with what size of an Agar sample to use when transferring to grain and just wanna poke a few brains to get some clarity.
How much of a transfer do you use when using Petri dishes? I started out using a sliver of agar (usually the most sectored and prominent growth) and now going back and forth between using sections to half or the entire thing cut into quarters.
Another question I have is:
The bigger the transfer the higher of a contam rate possibly, right? (What if the plate is fully colonized? Does taking larger samples still pose a bigger risk of a contam or is the puk impervious to having hitchikers since it has taken over the agar.)

Depends on what you're doing. If inoculating jars for colonization with a known clean plate, use as much as you want. I find 3 - 4 wedges the size of a hit of acid to be better than 1/4 plate, but others do it different.
If you're cleaning something up, obviously you want the smallest amount transferred possible.
And no a large wedge isn't contam PROOF just because it's colonized, if something else was on the plate and landed on top of it you could still bring it along in a transfer, say a mold spore or whatever.

Somehow it doesn’t seem to matter very much. I’ve used anywhere from a dime size piece to two entire plates. If you play it right with well timed shakes they still finish close enough together that it doesn’t matter ime.
Normally these days I drop 1/8-1/4 plate.

Basically the more you can transfer, the quicker it will colonize. As you're probably aware, make sure to look at the bottoms of any plates for hidden contams before transferring from them.
For my grain masters I usually cut a 1" x .5" rectangle on the agar, then slice that up into 10 rectangles. Scoop it up with the scalpel as a single piece (this can take a bit of practice) and dump it in the jar. When you shake the jar to mix it, it will break up into multiple innocation points.
