Colonization issues

Hey Guys,
I have an issue and hope someone can help me out.
My first grow I inoculated grain bags with MS syringe and they colonized completely in two weeks. The grow went amazingly well and I took clones and did agar work and the whole nine.
Recently I wanted to experiment and see which medium colonized the fastest and gave the best results when grown out so I inoculated the grain bags with a MS syringe, a LC I made From another MS syringe (I know that isn’t a good idea now) and the last with agar from a clone I took during the first grow.
So here is my issue. It has been 10 days now and NONE of my bags are showing any growth whatsoever. I even moved the grains around to see if there was some in the middle and still nothing.
The first go around my room was about 80 degrees and now it’s around 73. That’s the only difference.
I don’t see how the MS syringe, LC and agar all have failed so temp is the only thing I can think of.
Any advice? Should I wait longer? Can it take longer? I read if nothing happens in 10-14 days inoculate again but I also read it can take longer. The MS syringe and agar clone are GT and the LC is PE if that helps.

It is not the temperature, I can tell you that.
Maybe an issue with your grain prep?

I got all the grains bags off a reputable site.

Still nothing. Should I inoculate again or wait longer? I just don’t understand how none of these are colonizing even a little bit.

I can send pics when I get home but it’s basically just Rye grain bags that look like they did when I first received them. The only one that looks different is the LC one. The grains all look wet. I only put 2 cc’s in. The MS ones all got 2.5 cc’s and they look as dry as the day I got them.

All signs point to the grain bags being the issue, "reputable site" or no.
Unless you did something to kill or contaminate all three of your cultures

Yeah I was thinking that too. It’s the only thing they all have in common. My first grow was with the 5 grain bags and this time I went with just rye. Is it possible they are just going REALLY slow? It’s been 11 days.

For the spores, yes, but LC and agar should show something by now

Well, today is 2 weeks since I inoculated so I tried moving two of the grain bags around to see if there was anything going on inside that I couldn’t see. Still nothing. Maybe a wet grain here or there but that was it. I guess they are all fails. MS, LC and a whole plate of agar. Not one colonized. The site I got the grain bags off of is supposed to be like “The Site” for this stuff and 6 of their bars didn’t do anything. I really need to find a canner so I can start making my own bags and jars. So bummed right now. 😔

you're in for a lot of disappointments starting out. All I did last month was wash out infected BRF jars.

I just wish I knew WHY they didn’t colonize. I know the LC and agar were good because the agar was already colonized and the LC I put on agar the same day I inoculated and it’s growing out fine on the agar but not on the grain. The MS syringe was brand new, never opened, from a reputable place. The only thing was it’s about 6 months old. I did store it correctly and put 2.5 ccs in each bag so I did everything right, as far as I know anyway.
Just weird that all the bags, with different inoculation methods didn’t colonize. I guess there’s a sliver of hope for the MS bags but I’d think after 2 weeks I’d see a hint of something.
Sucks I’m gonna have to order more grain bags from the site since I don’t have a canner at this time and it’s really my only option right now. Maybe they just sent me a bad batch? I don’t know. Ugh!

Hey Guys,
I am pretty sure the LC and Agar grain bags aren’t going to colonize at this point but how long should I wait for the MS ones. Should I wait a few more days to give it an even 3 weeks or just get rid of them now?

If it has shown no sign of growth at all in that long I would just toss it. Bare minimum you should have a tiny speck of some mycelium somewhere. You should try to start up some more agar and then get a good plate and just do the blenderless LI and see how that goes. It's less risky that LC to me as you know the agar is solid if the plate is clean and even.

You've ruled out pretty much everything but the grains being the issue haven't you? Up to you but I don't see much chance of them doing anything.

Yeah. Seems like it’s the grains. It’s the only common denominator. I only asked because I read that it can take up to 30 days for MS to start colonizing, but I thought that seemed long. I have a few other bags but they are the same grains from the same company, so I’m nervous they are bad too. I have a bunch of LC that was inoculated from a third transfer that started with a cloned mushroom. I could try to use that. It grew out in agar clean when I tested it a few weeks ago so I know it’s good.
Just frustrated. My first grow went flawless and now I can’t even get a single bag to spawn. I wouldn’t care so much if I had a canner and was able to make my own bags and jars but for now I have to rely on buying them off the internet. I mean, it’s a reputable place but I guess things happen.
Thanks for the advice guys. I’ll probably toss them and grow out the LC on agar and start over in a week or so and hope I have better luck.

What did you do differently between this and last grow? What is stopping you from doing the grains yourself? If you want something done in this hobby, you are generally off better doing it yourself. I would recommend sticking to jars since they are easier to colonize. Learning from your mistakes is a natural part of the process of finding what works for you as an individual.

If you are buying bags off the internet from some vendor you should just take that money an invest in a PC. Making your own stuff is way better as hikeadellic mentioned, as you know what you put into the bag (grain/amount of water/ect) and you can rule out problems much easier than guess work around what they put in the bag.
