Recent experiment: tried to save popcorn grain from possible trich/green mold attack.
    #26967825 - 10/03/20 03:45 PM (3 years, 3 months ago)

Hi guys, sorry if this is long but will have a couple pics.

a sort of disclaimer. I have had some good luck here but I think it is solely luck. I dont know how often what I did will work, it may or may not work again. Could have been a fluke (most likely).

I was trying to colonise 2 jars of popcorn grain with GT spore syringe. My luck with grain is usualy bad tbh, yet I dont have many problems with contamination when it comes to PF tek style methods. The 2 jars were colonizing slowly, 1 then started to take off but very soon was very noticeably down with trich. Those pesky little green patches started. So I hoped the other jar would be ok.
The "clean" jar kept colonizing very slowly but had rhizomorphic bits here and there. Then one day i found a tiny green dot staring back at me.
I tried to think what could i do to stop it or save the good grains.
I tipped the grain out carefully so it was all stuck together, got a light and some tweezers and Inspected the corn cluster for green bits. To my dismay there was alot more green than I thought, BUT it was all quite isolated little bits (saying this, i am aware trich is basically everywhere once you see it in a jar, so I was less and less confident about what I wanted to try).
After picking through the whole thing, trying my best to separate good and not so good grains, I managed to salvage a very small fraction of the original amount.
Since it was likely already contaminated I decided to try a non sterile coir and perlite substrate from an old plant pot where the plant had died and the pot dried etc. Added gypsum, then I ripped up some oak leaves to mix through it since trich isnt so fond of material like that. I think I rehydrated the substrate with a diluted mix of chilli focus plant food in water, since I think I read somewhere that trich doesnt like acidic environment so much? Trying to give food that the GT would maybe like more that the trich.

I mixed the rehydrated sub with the small amount of grain in this small seedling propogator I have, lined with clingfilm (the tray had holes in the bottom so needed to cover them). Then put clingfilm down onto the top of the sub and wrapped it around the rim of the tray.

About a week or 2 later it seemed to be fully colonised, but not super white. Very mushroomy smell and no visible green patches.
One day noticed 2 tiny primordia much to my surprise, as quite frankly I didnt think much would come of this. I half expected it to just be trich riddled.

White mites: this is a surprise even more because it produced the below pictured fruits, yet there are a lot of these tiny white mites living in the sub. I did wash away alot of them at one point. They are attracted to excess moisture, but seem to live in the sub along side it rather than eating it. They are composters of some kind that dont seem to want mycelium or mushroom. Just moisture. I partly think they have aided this experiment, even if not they have not harmed it any like I worried they would. Anyone know what they could be? They look like round, off white circular dots. Perhaps beetle like but so small they are some kind of soil mite perhaps.
Also interesting that all of them were pointy capped, my previous attempts were mostly round capped.

Here are pics of what came of this experiment. I didnt take pics of the earlier progress as I didnt think I would be writing this thread:

Apologies for the black borders, had to take screenshots from my gallery to get them altogether. Maybe could have done it easier way but oh well

Well at least you got some fruits. It goes to show how hardy cubes are despiste everything you did wrong. I doubt you had trich in those jars though, but rather any other type of green mold. Trich is parasitic and would have ripped through the cube myc before it had a chance to fruit.

This was a "banda-aid" grow of sorts with a happy end but I'm sure you'll be able to produce clean spawn in no time if you keep at it.

I have successfully grown cubensis before in mono tubs spawned from PF cakes. This was merely an experiment to see if I could save at least a little bit of grain. I didnt think it would work to be honest haha. Obviously i would not recommend trying this especially if you are right that it was another type of green mold.
It could be that the 1st jar was indeed trich as that grew very fast compared to the jar I saved part of. Like you said, that tore through the cubensis myc. Perhaps i should rename this thread to "possible trich/green mold".

Either way surprised and happy with the outcome

Think I have only ever had 1 successful grain jar spawn, of ps. Cyanescens. Every other time my grains seem to come down with trich or other.

Yea and beautiful fruits by the way. Are you using agar?

I was surprised by that too, all pointed caps where in previous grows they were rounded. I have used agar, but these were actually from a spore syringe made from a print of a previous grow.
