Agar inoculation question

I will be making up my first agar plates tomorrow night, per Pasty's Easy Agar TEK. I would like to do all of my work at night after my family goes to bed, so that I can make sure all airflow stops in the house. So here's my question, is there any reason I can't just leave the plates in the PC through the day on Saturday and then move them straight to my SAB that night? There shouldn't be any detriment to them just sitting in there for a day, right?

I've done that before it won't kill them or anything although I can say the condensation will probably be a lot more as that's what happened to me. However you can flip the plates over and let them sit awhile then just open them upside down and dump the pooling water out before you inoculate them.

I can't speak to the condensation because i fucking hate those things and don't use them but, otherwise everything will be fine.

If you are going to use no pours pasty plates are viable but the holy grail containers are wayyyy better. However pasty plates are very useful for tiger drop and dropping the whole plate into LI bottle.

Eventually petri dishes will be the move however I just love the fact I can sterilize the no pours and whip them out whenever. I know i'm gonna love petri but I have not yet built up the reasoning to buy a shit ton when I have so many no pours available.
...That being said I do hate cleaning those little bastards.

you might be over thinking the cleaning. All you really need to do is scoop the agar puck out and reuse.

Yeah you're probably right. I always scoop the puck out and wash the dish and lids but since it's about to go right back into the PC that's probably unnecessary.. Good food for thought.

Unless something really strange has happened, it'll just come out nice and clean and the pc will take care of anything that remains. I'd only rinse them if there was something abnormal going on. Like if you accidentally dropped it in dog shit or something.

First of all, sorry for not responding. Posted before bed last night and just got off work.

When you say "sterilize and then whip them out whenever", are you saying that you store the already made plates in the fridge or something? I know Pasty's write up said you could do that, but I wasn't sure if I should make just enough to use. I have limited space so I will be doing at most 3-4 jars of WBS to Shoebox for now. But if I can reliably make a bunch and store them for later use, I would rather do that. That way if I get one with contam, I can do a quick transfer with an already made up plate, instead of having to make more. Also, when you take them out of the fridge, do you let them get to room temp before using them?

You can make no-pours and have them at room temp for months. I would say 2 comfortably but up to around 4 months or more if you pour them thicker. No need to store plate in the fridge unless you have a culture on there that you want to stall the growth until you are ready to use it.
