Doc9151's first Florida tampanensis find since Steven Pollock's in 1977 3

Here is an overdue log of a rare find that was sent to me by Doc9151 over a year ago of the first Psilocybe tampanensis to be discovered in Florida since Steven Pollock's single specimen was discovered in Tampa, Florida back in the late 1970's. Doc's finds were also the first recorded wild finds with actual sclerotia attached to a few of the specimens. I received the spore print of two specimens, labelled -
#1= Psilocybe tampanensis, location perdido river usa, #2= Psilocybe tampanensis location Beulah fl usa, this is (from) the specimens Alan did the DNA analysis on.
I got around to plating up several dishes from each specimen on the 14th November 2019 -


Noticed the first signs of growth on the 19th of November -

And these were some photos I uploaded of the plates on the 3rd of December -

Transfers were taken to fresh plates of some of the fastest looking growth from each specimen -

I used a small transfer from a dark patch of agar from specimen #2 to start a small jar of sorghum. The assumption was that this may have been the start of a sclerotia and I was trying to isolate some sclerotia producing genetics. One week later the jar had colonised and growth looked very clean so was transferred to a large spawn bag of sorghum on the 11th December 2019 -

I then chose multiple clean sectors away from the inoculation sites from each multispore plate of specimen #1 to a grain jar, and repeated for specimen #2 - to capture as much genetic diversity as possible from each specimen to be able to hunt for the best looking sclerotia from each for cloning. I made 3 jars in total... two from specimen #1, and one from specimen #2 -

Im aware of the risk of working with T1's of wild plates, but i had clean transfers on fresh plates as back ups and am just using these jars to look for the best truffles for cloning. For that reason using lots of sectors and capturing a larger sample range of MS genetics was beneficial for a range of sclerotia, and all growth looked clean. I also isolated away from the inoculation sites to minimise any piggy back contaminations from the point of inoculation, but all T1 jars colonised fine.

I noticed the first sclerotia forming in one of the jars after about 2 weeks -

After this I left the jars and spawn bag to do their thing in the back of a cupboard and forgot about them.
              
This is how things were progressing as of the 17 February 2020 -

Life got very busy and they were left to mature over winter and do their thing.
              
The extra time seemed to help as they really plumped up and developed a lot over the course of winter and here is how the bag is looking now -

The colonised grain jars are looking very mature and I am hoping to crack them open very soon to find the best sclerotia for cloning. I plan to grow out the best looking sclerotia from each specimen and then try and fruit each of those clones and take prints. This is how the grain jars are looking now -

I'll also try and fruit what is leftover after harvesting the truffles from each jar with the aim of getting some early fresh prints of each specimen for preservation.
I'll update with pictures as soon as I get time to harvest and take clones of the best looking sclerotia and with a bio-assay result.

I feel kinda silly knowing that a lot of mushroom names begin with the place they were sourced or someone's name, and that i never realized Tampanensis comes from Florida... seems so obvious now!

Wow wow wow.. like mentioned before. History in the making 
Cannot wait to see updates and eventually get my hands on some spores later Look at my avatar.. tampanensis loves me and I them 
Seriously tho.. sclerotia producers don't get enough love. It's so cool that we can get truffles and fruits in the same grow and the mycilium looks beautiful.. could save a grain jar for months until you feel ready to fruit 
Will be great to see the community spread these spores around the globe. Love this shit!

Sorry for the delay in updates, I had always planned on updating as I went along to keep everyone in the flow, but family life got the better of me and I failed to post my results as I went. Its been in the back of my mind ever since I finished with this project, my humble apologies especially to Doc1951 for the delay
Never the less, it was a very successful result in the end and I will upload some pics in the next week as I have recently had a change in employment status for the first time in a long time, and have some free time coming up. I will sift through the pics and upload them in the next few days.

Ok, here goes. The last photos of the bag and jars in the first post were on the 1st October 2020, so it had been nearly 11 months since first starting the project. A big part of the time was during winter and they were kept in cool, stable basement conditions the whole time.
Another month went by and with spring and summer hitting, things were looking good, some cracking inside the grains in the jars from sclerotia forming, and I noticed some bluing in one patch so decided to harvest before they got too old and warm again to avoid any rotting, plus it was perfect time to fruit them.
Specimen 1, Jar 1 on 28th October 2020 -
Specimen 1, Jar 2 on 28th October 2020 -

Specimen 2, Jar 1 (only jar) on 28th October 2020 -

Jars of each specimen were opened individually, transferred to a sanitised bowl and sorted to capture as much sclerotia as possible to find the best specimens for cloning, and the colonised grains were kept seperate to spawn to pastuerised coir/verm for fruiting -


Lids were loosely added to allow for gas exchange and the containers were set aside to colonise -

Here are some photos of the truffles from a jar of specimen 1 -


Four of the best looking sclerotia in this photo were selected for cloning -

And here are the results from Specimen 2 -
Again the four largest and best looking sclerotia were also selected from Specimen 2 for cloning -

Here are how the sclerotia compare side by side (specimen 1 on the left, specimen 2 on the right) -

And the two biggest sclerotia selected for cloning from both jars side by side (this time specimen 2 on the left, specimen 1 on the right) -

Lastly, here is the weight of the second jar from specimen 1 -

Specimen 1 yielded notably drier and denser sclerotia, and the mycelium had a darker hue of brown/yellow and had a lower yield than specimen 2 for both jars. Specimen 2 yielded sclerotia that were notably a bit softer/rubbery/less dense feeling and bruised easier.
The four clones from each specimen went to agar and were left to grow out -

On the 4th November 2020 the trays had colonised nicely -

So were cased with a coir/verm/potting soil mix that had been pasteurised, and placed into spawn bags with the tops only loosely clipped as fruiting chambers and exposed to fruiting conditions -


By the 5th November the clones were developing nicely and all clones appeared clean on the plates -

And by the 6th November were grown out enough to take some transfers to some slant style jars I prepared for long term storage, so I slanted the best 2 clones from each specimen and at the same time put the best clones from each specimen to grain masters -

On the 30th November 2020 I noticed the first pins from specimen 1, but specimen 2 decided to never to throw any fruit for the whole grow -

On the 3rd December they were growing nicely, and one fruit fell over as I was moving the bag, so it was harvested for photos -

This is how the grain masters with the best clones from each specimen were looking on the 4th December 2020, so I made up some spawn bags using sorghum and a sprinkle of coffee, and they were nocced with the grain masters in front of the hood -

 
The fruits were maturing nicely by the 5th December -

And by 6th December had started to open, so I decided to collect some spores -



The spawn bags were developing well as of the 7th December -

Lastly, here is a collection of pics of one of the other trays of specimen 1 fruiting, which was a few days behind the first tray (December 10, 2020) and swabs being taken -



Very happy to get some spores from specimen 1 to continue this lineage, but unfortunately no fruits came from Specimen 2 yet. I have a large spawn bag still to harvest from specimen 2, so will try again to fruit it this summer.

Last update for now was the harvest of the initial spawn bag that was made from a dark sector of Multispore mycelium from specimen 2 in the first run of agar dishes from the original post, which was inoculated on the 11th December 2019.
I noticed some contam growing on the surface of the substrate so it was finally time to open the bag, scrape off the contam and see what came of that bag on 4th March, 2021, 14 months after it was first inoculated -

But I'm a man of my word and I promised Doc that I would help continue the lineage, as it's such a lucky find in the first place and a huge privilege to work the genetics considering the story behind Pollock and his first and only other find from that area over 40 years go. Crazy shit.
I was always going to see this through though and had carefully documented each part of the process, and sclerotia are so forgiving and literally define patience - you can literally have them ticking over in the background and get to them as you need in and around work and family life. During this project I've had to re-roof the house during a cyclone, and went travelling for a month, and thats just a drop in the ocean - lifes been super hectic.
I still have some of the clones in spawn bags left to harvest, will update when one of those gets sorted.

awesome log!!! super inspirational!! by some miraculous miracle of fortune, a fortunate patch of "wild" tampas graciously granted me an audience; and delivered with them a few small fingernail size opportunities to form future fungal civilizations (spore prints). and after an undetermined amount of many hours accrued of neglect induced scorn from my spouse (i.e. research). im finally able to faintly wrap my head around the steps, procedures, processes, platforms, and media required to provide a suitable shelter for our fungal friends.
ive deduced that since id be starting from prints and working in less than ideally sterilized conditions and environments using just passable clean procedures, agar seems probably the easiest, most forgiving, least chance to fail route i could take as a self educated novice on the quest for stones. im actually planning on buying a pc sometime this week but am still trying to figure out what materials i can acquire locally and where to acquire them. something about ur agar instills a hope in me that i can do this lol. would u mind divulging some minor details on the ingredients and preparation of the media used for that agar and also that sorghum i recall seeing while out looking for other suitable nutritions that i wasnt able to find and am curious as to its preparation/sterilization or would i just follow a standard grain prep tek for sclerotia for idiots not using search correctly like me
