MSS to Agar?

When putting MSS to Agar is it best to use just a tiny drop and if so why? And is it better to inject multiple spots on the agar or only one ?

it is best to use a tiny drop, to minimize contamination. There are millions of spores in there. Just do one drop right in the center. make new plates for transfers.

Is there any tip/trick how to get get only a tiny drop out of the syringe? Usually for me it is hard to control it and have it only eject a tiny drop.

shoot a small bit out of the syringe onto the bottom of your sab. after you break the initial seal that can form in the needle, its easier to push individual drops out

Also, swabs can be very beneficial for spore germination... they absorb a lot of the excess water and you can run them over multiple plates to really spread those spores thin.

Yeah I quite like squirting spore solution onto a sterile swab and then doing a zig-zag pattern for germ plates
You can try to just put a small drop but I would more often than not squirt too much and then it makes a mess of the dish

I never had any luck with spore syringes to agar... they all went bacterial on me before I could even get myc to grow... swabbing was a game changer for me...

Thats how I do it. I also invert the syringe so that the spores go to the top of the syringe

Do you mean to use the syringe to wet the swab? When using a swab how would you get the equivalent of only using a tiny drop from a syringe?

Yes remember spores are microscopic. So a lil bit of solution with spores will soak up the excess solution so more of the spores hit the plate. I like to do a Z pattern. GL

No pour agar jars tek. Inject iso and flamed MSS into the FAE hole inside a SAB into the middle of the plate, then re tape hole with Micro pore tape. After a few days flip jars upside down.
If you wanna be a gangsta make a injection port with gasket glue or silicone on your lid as well as a FAE hole. (A bit over kill but up to you)

Apparently if you let the jars cool in the PC overnight it reduces condensation.
Slower temperature change over longer time I guess

I do that and still get condensation. But it only effects a small portion, but I am gonna start pouring it out in my SAB then use a sterile wipe to clean it up.

I have done no pour agar and gotten condensation but I let then sit around (not upside down) and eventually the condensation went away. I used pp5 containers (mostly Target generic or Dollar Tree containers).

Wouldn't that be equivalent to injecting a lot of MSS as oppose to just a tiny drop?
