Liquid Culture jar issues

Hey all, Just went to make 2 LC's from spores, and I had an issue that keeps popping up. I have been using plastic lids with SHIP's, 1-8 ports respectively, lol different story. There is also a 1 cm. hole in the middle with a tyvek filter on the underside of the lid. The jars themselves are 1/2 pint wide mouths filled with 150 ml. of H2o and 6 gm. honey/karo. This means the liquid is close to the filter/top of jar, and though I tested this last night with plain H2o in a dummy jar,injecting 3 cc.'s of solution in them today and liquid bubbles through a port or through the filter, and at that point I can only assume this isn't going to work out. I do it this way so I can extract the LC without opening the jar, as i use old spore syringes to extract. So is there a longer needle that I can find to fit on the syringe, or can I use a vessel that is larger,with more space between the top of the liquid and the lid, and just open it without fear of contamination? Do I need a filter on these even? Any help would be appreciated, I'm getting really frustrated with LC's and agar lately. Thank You.

I am not an expert but made a handful (or two) LCs myself which I have used with great success..so here are a couple thoughts which might help you out...
- spores to LC is a bad idea, since spores are rarely sterile. you mentioned agar in your post so I would recommend using Josex poke tek (biopsy method) to inoculate your LC - I am seeing faster growth with LME (2-4g/L) vs. Karo/potato flakes(2g/L each) but once colonized both works fine ime, honey actually (aahktuaalie) has anti fungal properties - you can use any size jar you want - LCs need less GE than you think - test your LC on agar before use - pour your LC (in SAB or in front of hood) vs using a syringe (it's kinda hard to suck liquid out of a closed jar plus you add a contamination factor by using a port) - yes, there are longer (3") needles - don't give up -> LCs are freaking awesome when they work out

Concise and to the point, unlike this post lol, thank you. Yeah I just started my foray into agar because I know LC's are unpredictable using MS solutions (Though when I DO get them to work, they are awesome). Tried live tissue in LC jar via biopsy, but no growth. And I don't want to pour LC in PF jars...Or do I (I use PF tek but with tyvek filters and SHIPs instead of traditional verm. barrier). Thanks for all your tips friend, no giving up yet!

Spores to lc is predictable
I've poured lc to pf style jars they actually suck it up nicely

Mushboy what you mean? What is this pouring? Do you makes PF jars with no verm barrier like me and then just dump like a little bit in and seal her back up? Thanks.

I made a LC from a PE clone onto agar. It colonized up nicely. Then I put it into syringes. Working nicely. The LC mix I used distilled water, turbinado sugar, and brown rice flour, I added a little food coloring. I inoculated 50 jars about 5 days ago, looking awesome. Almost ready for coir. I changed from metal lids. To plastic lids. I don't like the rust.

I pour LC to grain jars.

Some PE jars roughly a week or so after pouring.


I go agar>LC in media bottle> grain jars. It is my preferred method.
Mushboy has a whole thread on it...that's were I got the idea.

did you mean it was a liquid culture with flour? does it work like lme light malt extract?
