I have been kicking around the idea of growing some lions mane for a while now.  I went ahead and pulled the trigger on a grow kit.  I know for cubes, grow kits are a travesty, and a no fly zone.  I would never even consider doing it for cubes.  But I cant find any threads saying to avoid grow kits for lions mane.

We shall see.

I figured getting a grow kit would be a good way to jump into lions mane, and have a live culture to reproduce.

So what Ive seen from the research i've done on these kits, is its basically in a myco bag, already colonizing.  Once fully colonized, cut a few X's into the bag and let the fruits grow.

Here is the question:

If I cut the holes in a an SAB, can I use some sterile tweezers to pull a piece of the myc out of the bag and transfer it?

Assuming so, what would be best to transfer to?  a WBS jar, agar, or LC?

I would like to retain a live culture, and maybe even a spore print to add the species to my library and if I like them when they grow, I will use that to grow my own once its ran its course.

Any input from those of you who have done it before would be super appreciated!

You could try that, might have to clean it up a bit.  Then you could still clone a fruit if that fails.

Transfer to agar, you never want to transfer tissue of unknown cleanliness to grain or LC.

For your grow kit, make sure the bag is tight up against the substrate with no extra air inside it so your fruits don't grow in there, you want to force them to grow through the x's you cut.

Thats a valid point, i probably shouldnt mess around with grain, or LC.  I have read that LM will colonize WBS, which I have, so I was leaning towards a G2G. But especially with it being a pre-made grow kit, i probably shouldnt trust it enough for a G2G. 

I am a bit new to Agar.  I have been doing it for a couple months now.  I have not dabbled in making my own.  I have had extremely good luck with a premade MEA agar pack from Seaweed Solution Laboratories.  I just did a search, and it looks like LM will in fact colonize MEA.  Maybe when I get it in the mail, I will make a plate and transfer it. 

As long as it colonizes my agar, cleaning up a bit shouldnt be an issue.

Looking at the picture it looks like the block comes in a box that is missing one side.  Taking into consideration what you said about air pockets in the bag, i assume the box is designed to mitigate that issue for a novice cultivator.

Ok so i got my kit in the mail.    First question I have, is this colonized enough to fruit or should I leave it in the box it came in a little longer?

Next question is regarding the instructions.  It says to cut a 3 in X in the face of the bag.  I get that, but then it says Put it "X" down in a bowl of water overnight.

That sketches me out.  I know with BRF you dunk when you birth them, but Im afraid its going to be a contamination vector.

I plan to attempt to pull a live culture from the sub when I cut the "X" so I will be doing it in a SAB.  Should I put the bowl in the SAB too and leave it in there to soak?

I don't see the point of putting in your SAB for a water soak.  The only thing you're gonna be worried about there is a little bacteria in the water, which I wouldn't worry about anyway, and a SAB isn't gonna do anything to help.

I would do your best to get rid of any air in the bag though.

I will definitely do the best I can to get as much air out as possible. Do you think it's ready to fruit?  I did read that LM myc is isn't as prominent as cube myc but there are a few spots that don't look colonized.  I believe in the third pic, it shows a side that looks like myc hasn't fully colonized it yet.

That spot does look strange, a little too wet maybe, but either way I would fruit it :thumbup:

So the lions mane seems to be going well.  It's just starting to pop fruits out.  I took a culture of myc to agar when I cut the X into the myc bag.  Have done 1 transfer so far.  Kept the original dish, and it is fruiting!!  HH adorable little lions mane.

The picture kind of sucks. My phone doesn't like taking pix of things inside pasty plates.
