strange problem i had with my agar (no growth)

i used pre pour agar tek (found it on shroomery) and cultivated more or less (little problem with condensation) nice looking cultures with healthy mycelium which worked good. then i was away from home several months and didnt safe my mycelium and had to start from spores again.

but my agar didnt work anymore. i didnt get the fault because in my opinion i did everything like before. mostly i had not 1 mycelium growth in all the plates. only sometimes very milky looking myc. when i did a clone from a last surviving fresh mushroom the tissue spread its hyphae in all directions but no grow on the agar.

i suspected my malt extract could be to old (it got hard because i did not close it properly) so i got new malt extract. but still i had horrible results. now i changed my agar (no syrup addes in this one) and i did pay much attention to add the stuff into cold water and let it really cook for 2 minutes while stirring intensly.

now it seems that its working like it should again. i can see good white growth. so the last agar jars looking good again.

but im still unsecure what the actual fault was? are there brands of agar or additives which stop any mushroom from growing on it? maybe because u can store the agar longer if u use it for bake or other foods? do u have to stirr it very good when u heat the agar? what could be the reason i failed so hard before?

anyone experienced something similar?

hey man, glad to hear you got it going again, that sucks about before and sounds confusing.

imo question is whether you changed what exact batch of agar powder or other components of the medium were used, particularly when you started to get bad results. (and similar thing for when it started working again)
I'd ask how much time passed between good agar results and picking it up again with bad results, however i don't believe it had anything to do with that.

You contradict yourself or i should say some matters happen to be conflicting: it cannot really be due to additives because you first used old agar that got hard. I have to assume this is old agar because you used it in the past [with good result] and any additives would be in it at that moment as well. I guess you mean additives being the problem for the second new agar you got, however occam's razor says it tends to be better to look for simpler explanations: a single cause and not multiple.

I have used old agar before, yes it can attract moisture and clump and this can hinder dissolution. It takes longer that way. It is always better to put in cold water and stir up well imo even with fresh agar that works best. If you do not always do this, it can be more work to dissolve agar properly and hard clumped product would be an additional hindrance.

In any case in my experience the problem from this can be poor dissolution and esp if you are a bit hurried, the agar may crash out again when you PC it leaving you with unpredictably softer gel. This is more of a practical problem than that it should be a reason myc doesnt wanna grow in/on it.

I def cannot often a certain explanation but to me it sounds mostly like you had bacterial problems with your culturing that made it look 'milky'. Maybe endosymbiotic type bacteria are (symbiotic here in broad sense including parasitic) what can make them not grow, yet the contam may find it hard to grow on its own, idk.
the spread hyphae may have been your myc's attempt to escape without success.

Basically i do not know of something that could get into your medium randomly that would have such antifungal properties, maybe something like terrible pH but that doesnt come out of nowhere.

If you did not change anything either and got it working again, i would say it agrees with an explanation like the above.
If you changed what exact source of myc was transferred (different old plates, slants, or different new clone attemps) it suggests some were compromised and you transferred a contammed bit (can be harder to spot if you transfer from totally colonized plates).
If you did not change your myc culture source you transferred and got different results it suggests your technique because it is by definition the  main thing that is never the exact same, just as no moment is.
