Water/condensation on agar. Need help

Hey everyone I made some agar dishes using mason jars instead of Petri dishes and I keep getting standing water from the condensation that builds up during the cooking process and after I take them out of pressure cooker.

I used 5 grams powered agar 5 grams of some dry organic malt extract and 250ml of water

I pc for 40 minutes and when they get done I have lots of condensation that eventually turns into puddles... totally ruined my cloning process. I mean their growing but it’s blotchy from the water spreading the stuff around..

Can I just dump out the water during SAB work? Or should I leave them out in the open to dry up for a few days?

What am I doing wrong?

Yes I bring the 250ml to a boil then pour it into a dish to mix everything then dump that into jars, while still liquid they go into the pressure cooker

:confused: put the water in a pot and add agar mix in the pot and slowly bring the heat up and stir it while it heats up:thumbup: thats how i do it

Ok. I’ll see if that helps any. I think ima just dump it out unless someone says not to otherwise. Idk if I should add more agar or malt extract to help combat the condensation. After about a week they dry up to what looks like a normal plate. How long will agar stay sterile? Could I just wait till the excess water has evaporated?

Very frustrating

I would hesitate on dumping the water out. I’ve tried that before and the plates got very contaminated. I think it essentially gave contaminates a pathway into the plate.

Just wait for them to clear up. They will eventually do it on their own. Or just use them as is it shouldn’t be a problem

Arhghagrh. Dammit I was leaning on pouring them out lmao. I found 2 post with people saying pour it out. But what you just said makes total since like I can’t see away around it cause it would just be a river of contamination basically...

I got 3 jars in the pc for a test run on a fruit body clone. Ima dump one out, let one sit for 2 days then clone. And I’m not sure what to do on the last except leave it as a control seeing as how I didn’t change anything in the recipe

If you are using mason jars I would turn them upside down as soon as the agar has properly solidified, so the water collects on the bottom on the lid instead of the agar. Then just dump the water out right before you inocculate. That way you only open it once for dumping and inocculating and the water stays away from the agar.

I do 5.6g to 100ml of distilled water in a lab flask. Its malt extract agar. I prepare it in a pan before it goes into the PC, Add polyfill and a tinfoil top PC for 45 minutes. Take that out put it in lukewarm water for a smidge then transfer to cold water stirring it down the correct pour of 50c. That water could have come from your PC or you poured it too hot. I pour them in a sab then let them gel in there also. Contaminants are a real bitch sometimes. Wiping the plate down only risks adding contaminants. Unless you need the plates just run with it if its not contaminated and you added spore material. There could still be babies in that bathwater.

Nice! I turned them upside down and let the residual water set in the lid waiting for it to cool down to try n dump it one lid is plastic one is the metal style. We’ll see what happens. I got 4 more pcing right now to have more test samples

Ok I have another question about a technique.

If I leave the jars upside down and loosen the lids before tissue transfer can I take the jar off of the loose lid make transfer in that motion and then put the lid on After transfer ? Basically pull the jar off the lid make transfer and the put lid back on? That way I can dump excess water when moving lid to the jar. Idk seems like it might work what y’all think?

You are right.  Try to keep your movements deliberate and carefull, have paper towels down that are spritzed with ISO, and you should be good. I used jars, had the same problem, and that's what I did.
Try letting your jars cool, with the lids off/cracked before you pc. Then try and slow down your PCs cooling rate as much as possible. That can help as well.

Awesome!! Yeah I noticed when I rush things it’s much more wet than if I just let it do it’s thing. I got a pretty good feeling it will work but I always like to ask before I try something I thought up haha.

Are you using no pours? If so, just dump the water out before using them while in the SAB. Problem solved.

Have you had success doing this? I tried this method too in front of a hood and the plates contaminated on me. I figured cause pouring it out essentially provided a pathway for the contamination, but maybe I should try it again if it works well enough

Yeah I did it all the time in my SAB with no problems. Only with no pours though. I'm not sure how it would invite contams in if done right. Im not saying you're wrong. Just thinking. I would do it pretty quickly and with minimal exposure.

It's essentially the same motion as doing a tiger drop/tic tac toe drop of a no pour into a jar of grain.

Yeah I'm a huge fan of no pours.

Maybe I'll give it another shot cause that would be a simple way to get rid of those annoying pools of water. It's just last time a bunch of plates contammed up real bad and I couldn't think of any other reason that they did besides the fact that I poured out the water, and it provided a track for contamination. Maybe I was just sloppy that day
