What exactly IS an Isolate?

I've used the search function and I've found some answers, but I still don't really understand what I am looking for on my agar to actually transfer out an isolate onto a new agar plate.
From what I understand spores/ms syringes have many strains and the only way to get a good control of your grows is to get a single strain isolate and then grow out from there to determine if the isolate you got is one with a good set of genetics.
SO if what I've described above is correct then when I place some spores or a piece of mycelium onto an agar plate, it'll have multiple strains or sectors on it. What am I looking for to pick out a single isolate so that I can transfer correctly?

Don’t worry about that it’s mostly pointless unless you’re into ultra long term tedious projects.
Just look for the fastest most organized growth and transfer a tiny piece from the edge. Once you’ve got a decent looking clean culture the best thing imo is to drop it to grain, grow it out, and then take clones.
Isolate and monoculture are pretty confusing terms around here.
I’ve seen them used interchangeably to mean both a culture narrowed down to a single strain which is extremely rare, and then also just as a culture that shows consistent uniform growth in a perfect circle.

A good fruiting clone is worth 10 untested true isolates IMO. Most true isolates are crap and the juice not worth the squeeze. For our purposes a good clone is all you need.

A.K.A. When you say take a piece from the "edge" are you talking like the edge of the mycelium? Say it's growing out and looks like veins, I would cut a piece of the end/edge of the mycelium? Take that piece transfer until I get a nice "picture perfect" agar plate where it's a uniform circle?
None of my agar has ever done that I guess because I'm not grabbing the right parts when I do a transfer.

Yes. Leading edge just means the part that's growing the fastest. We grab that because it's potentially the most vigorous myc available. Transfer that to a new plate and watch it. Continue transferring until you're sure there is nothing but cube myc growing and then put it to grain. It doesn't have to be a perfect circle at all.
As stated, don't trip on isolating a single set of genetics. Just get good clean growth, put it to grain, spawn to bulk and then take a few clones. Grow those out and keep the best genetics on agar for future grows.

The only way I’d bother with true isolates is if I was developing a collection of monokaryons, then breed em and test those isolates. Otherwise a good clone will be all you need and give you more fruit in the meantime.
