Liquid culture

Hey guys,
I made liquid culture with a PE ms syringe a few months ago and inoculated two 3 pound bags. I read to use 7-10 CCs so I used around 7. The one bag grew a thick white mold all around it. It didn’t seem like mycelium. It looked more like a marshmallow consistency. I let it sit for about a month to see if any fruit grew just in case but nothing so I tossed it. The other one was just wet looking all the time and nothing ever happened.
My liquid culture has thick mycelium in it and I put a few drops on a Petri dish to let it grow out first to make sure it didn’t have any contamination since I am a newb and used a MS syringe to inoculate the Liquid nutrients to make the LC.
I think the issue was too much moisture in the bags maybe?
So, about a week ago I used the same LC and only used 2 CCs this time and the bag isn’t showing any growth at all. Just a few wet grains. I also put a few drops on a Petri dish again and it is growing on that with no issues but not my Rye grain bag I got from a reputable place.
Any ideas on what I’m doing wrong?

Well not necessarily. If you're going to make LC, use a clean culture from agar. Not a spore syringe. Also, sterilize your own grains. You're paying way too much to have someone do effortless work for you.

lc has to many contam vectors if u was to leaen agar and use a wedge to innoc a gm and them g2g is a way better way of growing but yes if ur going to use lc use a wedge then u wight as well g2g

Oh I know. And the place I’m getting it from always sends me 2 pound bags rather then 3 and I have to email them and they apologize and send me more 2 pound bags to compensate. Lol. But anyway, I can’t find a pressure canner right now. They have disappeared off the face of the earth. Lol.
I actually made another LC with an agar piece from a GT clone. It was the 3rd transfer, so hopefully that will work out.
I just don’t understand why it doesn’t work.

No. Lol. I bought the liquid culture solution already made off the same site. Very expensive for basically sugar water. I got the agar dishes off amazon. All I can do until a find a large pressure cooker

Well I need a larger one. Like a canner size. 16-23 qt range. Those are the ones that are hard to get right now.

Yep. Trying to learn so there’s a lot of trial and error, which costs money. Lol. But gotta do what you gotta do. For now anyway.

I actually put the rest of my agar dish into another bag and all the wedges stuck to the sides and it was super annoying. I had to cut a slit in the bag and drop them in then fold the slit and gorilla tape around it 😂. Let’s see if it contams out. Did do it in a SAB so 🤞

Some swear by it, some don't see the point. There's more than one way to skin a cat. A2G > G2G works great, and its pretty fool proof. LC is fun because you can expand it like crazy fairly quickly, but you can't see contams until you put it to grain or test it on agar.
I personally don't bother with LC until I have a solid clone culture that I want to expand.

I put it on agar twice and no contams. Just don’t know why it isn’t doing anything on the grain. How many CCs do you guys use?

I use 3-5 cc's per quart of grain and then shake it up.

A trash can.
But seriously, assuming it’s a 2 pound grain bag you likely want to spawn that to a coir verm sub at a 1:3 ratio. I would suggest a 40ish quart mono would be the ticket. Or make a larger mono and use both bags on it, however that carries more risk.

How many CCs do you use? I don’t know why mine isn’t colonizing. I also put an entire agar plate in one and the same thing. Nothing. It’s been a week. Maybe it just takes a while? When I did my first grow with a MS syringe it was like gang busters right away.

Yeah. But it hasn’t done anything in a week. I would think it was quicker than spores. I don’t know. I guess I’ll keep waiting.

If the LC is contamed or the colony in it lacks vigor it can be slower or an utter failure. If it’s axenic and I’m confident with it it will usually recover in 24-48 hours.

I put it on agar first to make sure it wasn’t contaminated since I used a ms syringe to inoculate it. I now realize that was a newb mistake but it grew out without contams in the agar. I shook up the bag today to see if there was any growth in the center and nothing but a few wet rye berries at the bottom. I don’t know. Maybe it’s a failure. I’ll just let it sit and see what happens. Tomorrow will be 7 days.

If the LC didn't recover after 3 days I dont think you'll see anything. Good news is you have clean growth on agar. Make a transfer if necessary or just chop it up and drop some wedges to grain. Heck, drop a little bit in some fresh LC broth if you want to.

My jars are opaque after 2 days of shaking my LC is raw sugar,honey, and BRF, food coloring.
