Pan Cyan tray that *almost* made it... what went wrong?
    #26942663 - 09/18/20 03:24 PM (3 years, 4 months ago)

I've been trying my hand at pans for a few months now without luck, but my last attempt is the closest I've come so far, and I'm trying to figure out what went wrong.

The relevant details:
  • Started with Pan Cyan Jamaican spores from a sponsor vendor, took to agar, transfered to get a clean plate, then used it to knock up wheat berries
  • On full colonization, spawned to pasteurized straw/manure/verm/gypsum mix (following the JOC Tek recipe) at a ratio of about 1:3 or 1:4, then let it incubate for about a week
  • Once fully colonized, I cased with pasteurized 50/50 mix, let it sit overnight, then put into fruiting
  • FC is a martha greenhouse with (a) a seed heating mat under a shallow tub of water (for temperature and baseline humidity), (b) reptile fogger that cycles on for a few minutes every half hour, and (c) FAE from a small fan that pipes in air from the top of the chamber and out the flaps at the bottom that are left open a couple inches (with the fan mounted to push air into a bucket with a few inches of water and from there through a hose, so it's not totally dry air going in)
  • I'm still dialing the GH settings between the fogger and the heater, but in general the temps range between 75 and 80 and the humidity between 80 and 95+ (based on an analog thermometer/hygrometer set in the middle of the GH)
  • I manually misted the casing a little bit for the first few days, but once it colonized, I just kept the humidity level high enough with the fogger that there were always droplets of water on the surface myc but never enough to pool

So the end result is that I got a bunch of pins, but they all seem to have aborted at about 1/4" to 1/2" in height over the past few days (first pins were 4-5 days ago but nothing has made it to maturity). And meanwhile, both the casing and sub have a distinct bluing that I'd normally attribute to dryness, but the casing has never dried out at all and the humidity has been constantly high. Looks like it may be a bit bacterial at this point too, but not sure if that's the root cause here or just the natural conclusion of the sub going bad for some other reason.

So here's what it looks like as of today:

Can anyone tell from these pics what the issue might be? I just knocked up 3 more pints last night so will be having another go at this in a week or two, so would like to figure out the problem now so I don't repeat any of the same mistakes next time. Thanks!

Oh, and should also note that the sub smells a slight bit off, but not distinctly moldy or rotten or anything, which is part of why I think it's probably going a bit bacterial (also the yellow spots, obviously). I haven't tossed it yet because the GH is mostly empty anyway and I want to see if there's a chance it'll recover, but I'm keeping a close eye on it in case it starts taking a clear turn for the worse.

It looks like you cased too early. That’s another thing I kept doing at first. Next time give it a few extra days to consolidate, it shouldn’t colonize the casing hardly at all.

If the surface always has water beads like that idk why the pins seem to be drying out. I guess crank up the overall humidity maybe.

Another possibility is the spores. There seems to be a lot of problems with screwed up genetics from vendors.

This is my first shot which was jams, way too dry so they only survived under the little wall. But it’s closer to how the surface should look during fruiting, mostly uncolonized casing.

Oh wow, yeah, the casing was colonized thoroughly. Might be genetics - I made an LC from the same plate and then knocked up some pp5 ziplock pints that were filled with the sub Asura recommends for bags, and they colonized great but then grew out over the casing in like two days after I applied it (enough so that i couldn't see the casing anymore, so I tried scratching it off a bit to give it a fighting chance). But in both cases I cased as soon as the sub was colonized - you're saying I should wait a bit longer then?

(And just to confirm, because this is what would happen to a nutritious casing - it's indeed just 50/50 peat/verm + lime and gypsum. And not sure if it matters, but I even ph tested it and it's right between the 7.5 and 8.0 marks.)

Yeah just wait like two days past when you would normally case it. If it does grow over it I’d just recase over it.

I did the same thing my first few times. Right when the surface was colonized I’d case it and then a day or two later it would be grown over and I’d have to do it again. I think it moves so fast the surface is covered before the whole sub is actually colonized so if you give it a couple more days it’ll finish and switch into fruiting mode and then it won’t run over the casing.

I use jiffy mix with a little extra verm, same stuff you listed.

Here's the plate it came from (forgive the condensation, it's been in the fridge for a month and a half):

Re: my sub and what it's fruiting in, see the original post. Thanks!
