Questions about strain isolation on agar

I have been doing quite some transfers on agar, and ran in to 2 questions:

1. The following image is Cubensis "Hawaiian" mycelium after 3 tranfers.
It has never been rhizomorphic so far, but sectoring now seems to be limited to 2 or 1 sector(s):




Is it fair to say that each of the 2 sectors can be considered being a clean isolated / monostrain culture?
Or can you not know so by just looking at the sectoring.

2. Another question about obtaining a clean culture;
I have B+ pinning on agar (after 4 transfers);

Does cloning this pin result in a monostrain? Or can that not be said for sure.
And if no, how do I know when i have reached a monostrain?

Any info/links on making single strains is welcome tbh. Am i correct
about wanting a monoculture for making slants? Or can i also use transfers
that are not yet singlestrain?

Getting a monoculture from MS will take dozens of transfers. And I'm not sure of this, but the cultures in your first pic are thick/fluffy/tall because there a very many genetics piling on top of each other

Cloning a pin will narrow the genetics present considerably, but, as I understand, there will still be some genetic diversity, tho not near as much as MS.

You do not need a monoculture to store on slants. You can store any clean culture just fine. :thumbup:
