Black spots

One of my jars has some black spots. I'm going to isolate it and spawn it separate from the rest of my grain jars. I'm wondering what they are? I highly doubt it's a contam, but I'm wondering what you all think.



The rest of my jars are fine, however, the black spots appeared after full colonization.

Are these cubensis? If so, it is certainly a contam, and I would not recommend trying to 'isolate it and spawn it'.

Just because I feel like I've ran into them in the past.
Now I'm certain it's a contam, the jar smells like ammonia or something.

Failure at any given step for any given jar. Improper grain prep, sterile technique, or equipment.

Or lack there of, more precisely. On top of whatever the black is, it has the “wet ice cream” look indicative of bacterial contamination and the fact that it’s not colonizing all the way is another indicator of bacteria. How’d you noc it? Either way, shit happens, even when using seemingly clean cultures on agar. Good technique come with practice.

I inoculated it with an Agar wedge.

I never shook that jar and I'm wondering if not shaking it could be the cause or somehow play a role for bacterial contamination.

Not shaking has nothing to do with contamination. It got in there through poor technique, contaminated culture, faulty filter or incomplete sterilization of the grain.

Not shaking it may have slowed colonization, which can give bacteria a longer time to get a strong foothold in the untouched areas. Especially if you did something like not shaking after a G2G and had an 'frosting layer' on the top and that's it. But the bacteria was already present, and that's the real problem of course.
