Does anyone do MS Syringe to Grain anymore?

Hello community, I've recently been struggling to get any WBS jars to show any signs of growth at all using ms syringes over the past couple months. I've tried multiple different vendors from the site and gotten the same no growth results from each syringe when put to wbs.

I know that agar is where its at in this hobby, but I had so much luck in the past few years with ms syringe to grain I never thought id have a moment where literally nothing I did would help the spores take to the grains. Funny thing is nothing grows in the grains even after a couple weeks or so. No contams that are visible and no bacteria thats visible.

This is what the inside of the jars look like.



These grains do have a bit more bust seeds but ive prepped some two weeks ago that had very little burst seeds but look the same and have no growth on them either.

There's little outlines where the seeds are, I dont think its where I inoculated with the syringe and the liquid just dried to the glass inside. I have another jar that was prepped the same method as this one but i didnt inoculate it. It looks the same as the ones that were inoculated. Same outline around the seeds but that could just be dried water that stuck to the sides of the glass.

I am at a complete loss and I have posted about this in the past or something similar so I can link the thread here if anyone wants to take a look at that as well to get a better understanding of the issue. I have put so much work into preparing the grains different ways to see if its my grain prep and at this point I dont think it is. The biggest source of confusion for me is, why in the past would syringes have worked so well when put to grain but now every single syringe I use on grain does literally nothing?

I do have agar plates and I do work with that so this isnt completely terrible but im so confused about whats causing this. As a matter of fact, this happening is what caused me to start working with agar in the first place. Still havent gotten the agar to grain tek down but im working on it and was wondering why the other jars that were prepped with syringes would do literally nothing anymore. Ive had grains colonize bacterial and still be able to fruit in small trays or shoeboxes and that was just about 3 or 4 months ago.

Im willing to just give up syringe to grain and stick to agar but Id love to figure this out first. Ive had several hundreds of great grows from syringe to wbs in the past few years but these past few months not a single syringe ( sourced from dif vendors) has shown any growth. It started off with seeds being more bacterial than normal and now like i said, no syringes will work when i put them to grains.

Agar is key to successful tubs IME. I’ve done one ms bulk grow, it turned out nice but once I moved on to agar I will never use a syringe to noc up a grain jar again. Definitely recommend you start using agar my friend,, you’ll be happy you did. ✌️✌️🤙

Ive already been getting the agar tek down. Just gotta work on agar to grain transfers a little more. Ive only done it 2 times but both were failures and contaminated with cobweb mold after about a week of colonizing.

Ill not be giving up on agar thats for sure. I just wanted to see if anyone else had been experiencing this issue or if there was something I could do differently to remedy the problem while I was still learning agar better.

Generally its not advisable to use MS syringe to oats or WBS.  Some vendors do provide clean syringes, but never count on that. It may take over 2 weeks for grow to appear on agar.  Agar being a flat surface is going to be much easier to spot and confirm growth.  While oats, or bird seeds are 3 dimensional and the growth is going to be harder to spot, and may take longer to be seen on the outside.

Since your using a new vendor, and having problems; I would suggest starting on agar.  Not just to try and clean up and isolate it, but to confirm the viability of their product.

Not all spore vendors are the same, and some vendors appear to use very dilated liquid cultures with sterilized spores; and I've had some spores germinate in the syringes themselves.  Needless to say growth is much quicker from those.

If their is no growth on agar, or your oats/WBS I would highly suspect the syringe.

If your able to grow on agar fine, and it dies out or doesn't colonize the grain/WBS that would be a fun issue to troubleshoot.

Thing is that very situation has occurred, you can see in the other thread I’d posted a few months back that I did manage to get a completely clean agar plate, actually got a few and when I put it to grains via tiger drop, they colonized a few grains then stopped and cobweb started at the top of the seeds. This is likely due to poor tek during the transfer of the agar to the grains. I will practice much more with agar and get cleaner with the tek.

I’m going to assume my grain prep is fine because I’ve used several different methods but every method I’ve used in the past resulted in growth while everything done with multiple different vendors syringes recently just seems to do nothing.

Tomorrow or tonight I will prepare more agar plates and see how things turn out with the most recently purchased syringe. If it grows on agar while doing nothing on grains then I’ll try to figure it out even more. I would assume it’s easier to germinate on agar than wbs though. So that may not be the best test to do trying to see the viability of the spores and my grains.

I did MS syringe to WBS a couple months ago. Had a couple cc's left in an old APE syringe so I put it to a half pint WBS then did grain to grain. Worked great. I use FooMans WBS prep. Haven't had any problems with that tek so far

I don’t. But simply because it’s so easy to get a plate going, and I’m never in a hurry.

I’ll attempt one more syringe put to grains tomorrow night and I’ll use the foomans tek. I’ve never tried that one before.

Hoping to get something going with the wbs but while I wait for growth on wbs I’ll also work more on agar like I have been. Just gotta get the agar to grain transfer down a bit better. 👍
