Why does agar melt in the petri dish?

Hello sir. I made 12 petri dishes using agar agar on 06.09.2020. I dropped spores with liquid water into petri dishes using a spore syringe tek. Today, 3 of them switched from solid form to gel, jelly-like form and I threw those petri dishes into the bin. What is the reason of this? My room temperature is 79-83F and I store them in the dark.

250 ML Potato Broth + 3gr agar + 3 gr sugar powder. This is the recipe which i used.



What do you think about theese myceliums, are they healthy? Thanks.

You need more agar. its liquid probably because there just wasnt enough in there to solidify it

Mycelium looks good in the pics though. kinda hard to tell with the quality

Low pH can cause issues with agar gelling. Also some species of gram negative bacteria can break down agar, reducing it to goo.

Take from the more robust and vigorous colony. See how it does when you do. Take lots of transfers and throw most away. Only keep what’s good. If nothing is good start over, don’t waste time chasing stuff that isn’t there.

Sir how can I strain the potato mix? I cannot use a coffee filter because the water does not drip at all. I need some clarity.

Unless you want to build a contamination collection :frown:. I store my busted plates in a case so I can collect every disaster for future reference. All three of those samples look fine the one on the left has good growth.
