Using refrigerator stored petris
    #26932397 - 09/12/20 07:35 PM (3 years, 4 months ago)

When I read about storing colonized perti dishes it seems you can store them in a zip-lock bag in the refrigerator for months at a time. But I have also read about not using them in grain spawn if refrigerated. Two questions come to mind:

1. What are refrigerated petris good for if not spawn use?

2. Do you have to let them warm up before use and if so, how long?

Much thanks!!

They're good for spawn, transfers, masters, whatever you want to use them for. The only thing that matters is the mycelium still has to be active (not dead). Let it sit at room temp for about a day before using to get it going again. This is optional though, it just gets the mycelium running before you put it into the jar. I have inoculated jars with cold mycelium before and it just takes about 48hrs to see any movement.

I like to use them as soon as possible. I use master slants for storage.

Refrigerated perris are good to use for anything, assuming they're clean. Where you can run into a problem is if you unknowingly contaminate them before putting them in the fridge (or they pick up a contam in the fridge somehow) and then use them straight out of the fridge.

The contam wont have enough time to make itself known on the plate, thus you'll see a perfectly good plate. Once it gets to room temp and on some grain, the contam will do its thing. So that's why if you're not sure on how clean your plate is, you either wait a while after pulling it to check for contams or you make a transfer and use that transfer plate on some grain.

Hope that makes sense. If not, let me know and I can clarify.

IME refridgerated petris end up with big pools of water even if the plate was condensation free initially. You can store in an insulated cooler to avoid this, but I just make slants I want to store stuff long term. Its more robust.

Makes perfect sense, thank you!
