There's several issues to address here, let me do my best...
If you took a lion's mane from the wild, and cloned it, that would be a culture. You could name it LM2000 or whatever.
If you collect spores from it, you are not getting that culture. Those spores you collect will produce different cultures. So no, collecting spores has nothing to do with saving a culture.
If you collected spores from your Sporeworks culture of pinks, and grew them, you could do whatever you want with those cultures. They just won't be the same as the culture you're growing now. They'll be genetic luck of the draw, maybe good, maybe not, but not the culture you're growing now.

With these pinks above in my pictures, I colonized the grain spawn, and then transferred the grain spawn to 5lb substrate block, and they colonized in 5 days.

Really nice! I got the syringe from sporeworks and inoculated a 3 lb grain spawn bag I got from Midwest Grow Kits. I'm about 4 days out from injection, so I have some weeks left before transferring the spawn.
Where did you get your substrate and how exactly did you make the transfer?
That's awesome you got your pink oyster syringe from them as well! I love midwest, I haven't used there grain bags, but alot of other products! Out of curiosity, with it being almost a week since you inoculated the 3lb grain bag with that syringe, how is it looking!?
But to answer your question on the next step, when your grain bag is fully colonized.. I started to buy the 'masters mix', it is 50% sawdust pellets, and 50% soy hull pellets. You can find the masters mix in bags online for a decent price. With the 20 lbs bags I bought, it gave me a recipe for 5lb fruiting bags, 5 cups (2lbs of the pellets) and 1.4 liters of water (3lbs. You will also need bags with the filter patch, probably similar to the one your grains are in. I use 5-7 lbs bags from Myers mushrooms. You simply measure out the 2 pounds of pellets directly into the bag on a scale, or cup measured, then you add the water directly in the bag, the water will absorb the entire bag within a few minutes. I dont mix it that much at all now, unless there is a dry spot. You then need to sterilize the bags, by pressure cooking them for atleast 1 hour and 30 minutes. After the bags are all soaked, I crease the top and side of the bag down to release all of the air, and I tuck the top of the bag down the side/to bottom, and fold about an inch and a half or so at the end of the bag, once in the pressure cooker this will make a temporary seal.
After the bags have steralized, and cooled. You need to have a very sterile room, hopefully you have a still air box you can work in, or a flow hood. You can open the bags, and use about 4 oz of your colonized grain spawn, or a cup, or even alittle more if you want. I believe on some mushrooms adding to much can mutate the mushrooms. I like to use 1 cup of grain spawn in my 5lb bag. Once you get the spawn in the bag as quick as possible, you can impulse seal the bag, or zip tie the top, but make sure you have air in the bag. After its sealed, shake the bag really good, making sure you mix the spawn around.

Those spawn bags aren't moving much at all, which is unfortunate. Not sure if it's the room I have them in or what. I just have them in a corner of a closet, where the temperature moves quite a bit, from maybe 70F at night to 77F during the day. Seems like a decent range for colonization, but maybe too much swing?
I have been getting into agar, and there's no doubt my Pink Oyster spores are slow spreaders. I have a Blue Oyster CULTure on agar that has almost overrun the plate, but my Pink Oyster plate only has two small dots starting. I inoculated both plates on 9/12, so five days ago. Both syringes were from sporeworks.
