Just starting agar!

I just started growing, and decided to get into agar!
I started with pasty no pour, and made 12 120 ml glass containers with it.
I'm about to inoculate for the first time and I was thinking how many should I start with, with my first inoculation?
And second question is, when to make the first transfer. I have read that I need to make my first transfer as soon as I see growth, to a new plate? And do I make multiple plates, for my first transfer?
I also read that the cleanest part, when the mycelium is growing, is its edges, because mycelium grows faster than contaminants, true?
Just looking for a little bit of advice regarding agar just used a MS syringe for 20 jars, but wanna get my agar ready with a clean culture as soon as I can, so I don't risk making some bad spawn. Love it already

Start small... i was always worried about contams so I would do 6-10 plates and then take a few transfers from each... before too long I had a whole tote full of plates... unless you're super messy, your contam rate should stay below 10%.
First transfers can be taken anytime... some people advocate for early transfers but I tend to let them grow about 50% of the plate before I make transfers. I like to see how the myc behaves for a little bit.
As for the myc growing faster than contams, I've never seen evidence to support that. Also, different contams will make the myc react differently. Again, thats why i like to let mine grow a little longer than suggested. The leading edge is usually farthest from whatever contams you may have brought with on the initial transfer.

Okay thanks Buddy, I'll go and inoculate 4 i guess, 4 each time, since I have 12 in total. And I'll let it colonize for a bit before making the transfers.
I just opened my PC and micropore tape was wet. Do you just let it dry or should I switch it?

Don't switch it! Lol when that happens to me I usually let them dry before I inoculate and once they're dry I throw another layer over the first one. I actually started doing the second layer of my tape on all jars just to help prevent contams.

yeah... i had an issue where after pc and inoc they'd be cruising along and out of nowhere there would be a spot of green on the top... my hypothesis is the pc degrades the micropore tape to a point where the pores are no longer micro... lol so the second layer was originally done as a test and I've since had no more random green spots... and mp tape is cheap, grain spawn may not be financially expensive but thats a hell of a lot of time to waste... just my 2 cents.

starting with no pours is a great way to get into agar and ive made my share of pastyplates over the last few years however the only thing about them is u dont have the room you do in a pitri and its easier to put 1- say 9 transfers on 1 pitri to save on plates for a short time like when taking germination points from a streaked plate for example. but usually i wont grow my cultures out very big even when going to grain so i can still even get 3 cultures on 1 plate no problem. its not that important it just helps to keep from always having to pour is all. but i would use the no pour pastys for as long as your comfortable with them and after a while if you feel you would like to try to pour some go for it. it def takes some practice tho so the sooner you learn the better off ul be. but il still even make pastys once in a while if im outta plates or if i dont wanna waste a plate say for somethin like tryin to germ an older print that most likely wont germ or if i only wnna make say 1 or 2 of a pda or other recipe than my standard mea. my point is tho they are both valuable tools to have available when u need them so just practice one and then learn the otherπππ
