Multispore to agar vs cloning
    #26911955 - 09/01/20 01:12 PM (3 years, 5 months ago)

I have dont a lot of multispore monotubs. I have agar and spore syringes. Does it make more sense to do a monotub or shoebox from the multispore and clone it, or to put the spores on agar and isolate until noc'ing jars?

Despite having success going spores > grain > clone on agar, my opinion is that you should do agar first.  There's just too much work that could be potentially wasted otherwise.

Now, that doesn't mean you have to try and isolate something (other than just clean mycellium) right away.  You can put spores to agar, make sure it's not contaminated, then toss that plate in a jar or two.  Alternatively, just let the plate fruit and take a clone from that.  There's all kinds of fun to be had.

If it was me I would put the spores on agar, transfer until clean and use that. No need to try to isolate, just get clean mycelium and inoculate.

I guess I have always had luck with clean spores. Is the only purpose of going spore to agar to make sure its clean?

it being clean cleaner cleanest is the most important thing in the hobby, so its "only" purpose is one of the most important things overall.

its either to clean up, or to isolate

Yeah I usually do to but some strains are more picky than others. I usually only use agar for panaeolus strains. It's up to you and depends if you want to isolate or clone.. I don't trip very often so I usually just use spores.. we're all in a different situation... It's best to start with agar though....

Isolating MS on Agar won't ensure you get a heavy fruiter or fast colonizer. The only reason you're going from MS to Agar is to ensure cleanliness prior to inoculation. If you don't care then just inject straight into jars/bag, produce yourself a fruit and clone. From there you can work on isolating.
