What is on my monotub sub
    #26904855 - 08/28/20 07:52 PM (3 years, 4 months ago)

So for a little background on this tub. I used a ms syringe of gt to inoculate a wbs jar. Once it fully colonized went g2g. Let those colonize and mixed 8 wbs jars to 10 coir/verm jars. Everything has been within the time limit of what a jar should take. Everyone colonized In less than 2 weeks, smelled really fresh and everything has been lookin great. Well i spawned this tub 2 n a half weeks ago took one week to colonize and then I let it sit for a couple more days hoping for pins but didn’t see any so put it into fruiting conditions, so it’s been one full week in the fruiting conditions and I have 3 small pins on one tub and a lot of primordial growth. But I have been slowly noticing a white patch forming on the ends of this tub and another tub  directly underneath it. Everything was fully colonized even tho the picture dosent really look like it. It was super white until I misted it with the spray bottle then it kinda went to what it is now. I don’t have any standing water on the substrate itself just light misting

For my grow box I have a ac unit inside to keep temps down and it’s sucking a lot of air on the side that this growth is forming. It has a slight gray discoloration in its worst spot but everything smells like fresh mushrooms. I mean dank I can smell fresh mush whenever I’m in the other rooms after I open a tote. So is this some kinda trich? Or is the air rushing past these walls causing my substrate to dry out and this is the result???? I’m losing my mind over this and cannot figure it out please help

Is this the cause of the tub being too dry from the portable ac unit pulling air across the tub or is this contaminated bacteria?

It looks like a patch of bacteria. Did you do GT liquid culture or did you use a spore syringe? Spores to grain does not have a great success rate in most circumstances.

If you are noticing pins forming (if it were me) I would allow it to continue to grow to get some fruits. However in my experience if the substrate is looking anything but white mycelium the clock is ticking until it turns into mold and the whole tub needs to be trashed.

Keep an eye on it and hopefully you will get some fruits but I think it is only a matter of time before you will want to pitch it.

More experienced growers might have better advice but at my (noobish) level thats kind of what I would be expecting based on the pictures/information shared.

It was a multi spore syringe to wbs, I have done it in the past with just a syringe before I ever got into agar and had decent success. I had some agar going but the wbs looked too good not to g2g and it all looked and smelled great but it’s still a man syringe .

What I can’t shake is the exact same patch is forming on the tub directly below this tub in the exact same spot. Whenever the ac kicks on it pulls a lot of air across this side of the tubs. Idk what a dry monotub looks like personally so idk if it’s dry or bacteria or trich or what?

Can I clone a pin is the pins I have don’t mature?

I think it’s bacterial something in that spot but it also looks like mold is about to pop up on the right side and the left looks pretty unhappy too.

The liner is blocking but normally when there’s a shadow like that up top you can see worse on the bottom.

That’s what I’m afraid of. It has lots of primordial dots under the white patch growth but idk. What does a dry substrate look like? Cause the pics i see online are Similar but then again this looks to have white moldy conditions too.... idk I just wanna have this work.... fml

Aside from the contamination stuff the myc looks like it’s been getting soaked and dried out so I’d ease up on air and misting and just try to keep it more humid.

Yeah I have a reptile humidifier on the way but it has been over 3 weeks and still hasn’t shipped. I believe it’s a combo of dirty Multi spore syringe and what you said wet then dry wet then dry.

The tub that has pins I wanna see what they do. I would like to clone a pin that way I can skip some agar steps for time. I had some agar going but it got put in icebox for storage. Mycelium was very wispy and no rhizomorphic growth so to speak.

What’s the best way to clone a pin? Should I take the pin from the substrate and put it I a jar/ baggie for the SAB?

You don’t need a fogger for cubes, just gotta learn to dial in the tub. If it’s drying out it’s getting too much air so tighten up the poly. It should be holding water on its own so there’ll be no need to mist it either.

For cloning it doesn’t have to be a pin. Bigger is easier to get clean tissue out of once you rip it open. You could throw one in a bag if you want but I’d try to wait until everything’s ready to harvest the clone.

Ok sounds good to me. I’m gonna chalk it up to it being multi spore syringe. I have a agar dish and a wbs as a control group and I haven’t seen any growth between the wbs and agar. I only opened the agar and wbs jar in the sab to simulate a g2g or a agar transfer. So far so good on the control so I don’t think it’s my sterile technique but then again here I am with a contamination so idk.

It’s not rare to find bacteria in a spore syringe, and then g2g a spore nocc’d jar makes it even worse cuz it’s just more time for the bad stuff to get buff.

I’d put some spores to that agar and roll on from there

Indeed!!  So here’s where my ocd and fears come into play.

How assured is agar, say if you get a clean rhizomorphic growth Sector to wbs, will that be the ticket provided the rest of the process stays up to par? I’m running out of problems to focus on and fix, temps are correct I sterilize my wbs at 15psi for 2 hours 15 minutes, very clean calm SAB work, idk seems like I’m cursed hahah

Yeah that’s the way to mushrooms. There can be tricky hidden contams on agar sometimes. A culture can have something nasty meshed with it that you can’t get away from but that’s not very common.

Just germ some spores and transfer a few times until you’re getting totally clean plates and then drop a few wedges, and try blenderless LI. Once you see clean myc in a few varieties and know what you’re looking for it’s really simple, you just have to stay on your game.

Big facts! Ok that’s what I’m thinking cause I mean what else can you do really. Granted the rest of the process has to work.

My problem is I grew lbs with just a multi spore syringe Years ago and I’m guessing my arrogance of ohh I can do it just like last time no problem. Hahaha nope back to the drawing board

My biggest problem when I started agar was dropping wedges in. I had never lost a jar of spawn doing syringe to grain and then my first agar inoculation contaminated 3 out of ten.

Wow so you get successful flushes more so off the syringe than the agar? That’s confusing lol

Lol I guess technically yeah cuz I’ve got a perfect record with syringe to grain.

I got better at dropping wedges pretty quick so it’s not nearly as bad now.

The tubs from agar are way better though.  plus you can’t clone from syringe.

Ok so a update on what’s happened. The top tub that had the gray patch now has a small pin and their are a few more little pins threw out the tub. The tub with the pins in it is the other one I was talking about. It has the same kinda white patch in the same spot as the top tub that’s what made me think some kinda damage from the ac pulling air across that side of the tub.

I still think it’s a weak multi spore grow that’s got problems but why the hell is it pinning in that patch of gray? Lmao I had some trich contams years ago and that was blatantly obvious but I cannot figure out what kinda contam this might be that way I can try to source the problem better. Idk what you guys think.

Those pictures make it look a bit better off than the first set of pics. It still doesn’t look healthy but from the first pics I assumed mold would annihilate everything post haste but it’s still truckin.

The myc network will always try to reproduce and contamination can even cause early pinning sometimes as a last ditch effort. I really don’t know what’s happening here though. The whole thing does look dry, or like the myc was allowed to get too dry and go leather at some point, and then went grey or sum’n.

I see now those are two different tubs. Idk yo. Print those things and swipe to agarasourus.
