Hard lessons learned for sterile procedure
Starting from scratch
PP5 MS following Violets selection process first vs learning the hard way. Not all fast pretty mycelium fruit. These clones will see agar then grain and shoe box. Man starting over sucks but maybe one of these will produce better stock.

This one I did not have to go all the way back to MS. Pulled a slant but I can quite tell if these have that contaminate that destroyed almost all my work. Maybe I’m just paranoid now.
“Second verse same as the first” These clones will see agar the same as the GT. Grain, shoeboxes, clone the best genes.

All the way back to MS. The syringe was definitely dirty. I was surprised this failure even produced. While teasing out the top by casings to prevent blobs i went one layer too deep. Two 1/4” was enough. I pissed them off and drove them to the sides. Or so I thought. After removing the casing layers where I ruined the top I roughed it up a bit and pulled it from the shoebox to let it pin out from the sides. Then I saw hope for the top layer! One week later trich took over. Of the 5 clones only one looks promising. Rest were contam.

Making some plates there was a bunch of shit that had already coagulated at the bottom of the media bottle when I went to pour which is frustrating we'll see how they go I ordered some activated carbon/charcoal from Australia and it arrived a few days ago so I was anxious to pour some black plates.

10 grams agar agar 10 grams LME. 1 gram activated carbon/charcoal PC'd for 20 min Yadda yadda
I think I'll whip up another 20 tomorrow and see if I can figure out how to avoid it all gumming up at the bottom without getting bubbles, and being at the right temp to not get condensation :/ IR thermometer would come in handy rn.

I use some white metal mesh thing for drying dishes.
I want to take this opportunity to say, boo yaaaaahh!

Pink teeth for the win.
And some natalensis pins just kinda emerging from the myc rubber. I had to peel some off to see them.

A little bit o dat PE6

And I had two questionable qts so I threw them in together in this 15qt sterilite. I really like this thing and will use good spawn next time, probably 3 qts. I got an unsurprising visitor this go. There’s more about to pop up in the bald spot diagonal from the first patch.

I swabbed a few chunkers from my first APE and they be germinating quick.
