Isolated genetics agar mono
    #26902650 - 08/27/20 04:20 PM (3 years, 4 months ago)

So I am trying to find "clean" growth away from any contam, any input is appreciated.
These are 500ml h2o, 10g agar, 4-7g lme plates poured and transferred in front of my flowhood.

Here are 3 plates that look like isolates to me
They were cloned from a fruit body, and transferred 4 times.

Can you tell from these pics if these are clean from any bacteria/mold and are truly one isolated genetic colony?

sorry about all the blank spots, had to burn my fingerprints

They look clean but not isolated down to one strain. It’s pretty much impossible from what I’ve seen here without using a microscope to put single spores on a plate.

One thread a tc went over a hundred transfers without getting an isolate. A lot of people are starting to think random mutations will make isolate change quickly even if you do manage to get one. Pretty deep into the project the guy would get what looked like an isolate and transfer it to make sure and it would look good for a plate or two and then suddenly sector again.

You need to get pretty lucky to end up with good genetics when you narrow a culture down a ton anyway.

Thank you very much for the info a.k.a
Is there bacteria/molds that you can only see with a microscope? like ones piggybacking along for the ride.
Or can you generally assume once you get uniform growth with no odd colors/spots its clean.
