Hi all,

This is my first time using agar. I nocced up some some plates of PE6, PESA, GT and Natal Super Strength using LC I bought from a vendor.  Apart from the GT and PE, most showed a milky white contam. I transferred wedges to new plates and the pictures you see are of these new plates.

They all look pretty good to me, contam free unless I'm missing something.

My plan is to transfer again, just to be sure they're contam free and also practise a little isolation technique, choosing the right section to transfer from. from this second transfer I plan to do a final transfer and then use those plates to inoculate grain jars.

My questions are:
1. should I wait for these to grow out a little before transferring? The NSS and GT have some good rhizomorphic growth while the others appear tomentose. On my GT plate that I transferred from and which has had time to grow out, there is good rhizo growth, so this would suggest waiting for the plates to grow out a little and the rhizo starts to show?

3. Can/should I be cutting wedges out of tomentose sections instead of sections with rhizo?

3. Although there's only one PE plate in the photos I have another 3 identical looking PE plates. I also have 3-4 plates of the other strains. Should I do a transfer from each plate and then choose the best one from those plates to transfer again Or just choose the best one and do a couple of transfers from that plate ? My understanding of what to do in this situation is a little hazy...

It looks like you have clean cultures so the answers to your questions are mostly subjective. But here's my perspective...

1. I typically transfer when the plate is 50-75% colonized.
2. Rhizo or tomentose... Between the two I haven't seen any huge differences in yield or potency. Starting from MS, I usually select the sector that grows most vigorously (although it does not always correspond to yield, potency, or whatever characteristics you select for). I've also seen myc that changes growth habit after prolonged storage, changes in agar nutrient content, etc.
3. Choose whichever looks more appealing. It's all up to personal choice. Once you fruit, you can select favorites to clone from. The clone generation should be more consistent.

Cultivate, observe, select, repeat. Don't worry about making wrong choices, you have a lot of genetic material to select from. Keep spores in reserve and you'll never run out of options to explore.

I dont like the third plate. I like 1,2, and 6 mostly.

1- ITs better to transfer earlier than later if you are still cleaning the culture.
2- Just take the leading edge, for me it has often been the rhizo that is leading.

3- dont know really. In the end what grows best in the box does not always correspond to what grew best on the plate. ITs a hobby, take time and see what you liked. Always keep a backup of your plates just in case. After the flush you can clone what you like and work with that.
