Can you deposit a spore print directly on to an agar plate?

Thats the idea. Either run a sterile inoculation loop over the print, scrape with a scalpel, or use a sterile swab to get your spores onto agar.

Or do you mean print directly onto agar? Not sure how you would achieve that without contams.
Also a waste of spores. Print to foil and only use what you need.

I mean just put a cap on a plate, spores drop and remove the cap. Probably would get contamination tho, just curious π

Bad ideaβyou're covering the plate in spores. Besides that obvious problem, the idea is to scrape absolutely as little as necessary, so you're not picking up contams. The more you deposit, the higher likelihood that you'll get bacteria or mold along with your spores.
Stick to Genghis's advice... unless you wanna run an experiment and show us!

Maybe later. Inner stem tissue transfer seems more viable anyway. π€£π€£π€£

You can print a cap in an unused (no agar/medium) sterile petri if you wanted. That way you can admire it without having to open a foil in the SAB, or compromising its cleanliness in any way. You can also monitor how much is left without opening it.
I thought about doing this, but never did. I like the much smaller form factor of traditional foil prints.

Put a dab of Vaseline on the lid of the agar plate and the cap onto the Vaseline so it will stay suspended above the agar so only the spores land and the cap never touches. After printed cap off the dish with a clean lid.

I've seen people use plates to collect spore drops. They didn't have any agar in the plate though. They just took a proper sized cap, threw it in and closed it for a day or two. Opened it back up and pulled the cap out and just resealed it like he would a regular petri dish. Like I said though, he didn't have agar in his spore print petri dishes.
