HELP pls! another "mold or mycelium" question

Hi there :smile:

so instead of doing another round of "cleaning up" mold I am seeking help in the community...
those are all T1s from MS plates. non of them look organized and/or like mycelium to me.

what's your opinion? toss or keep transferring?

also.. is there any info out there that LME agar causes weird looking growth? I have done a lot of plates now but non look "really" good :frown: even the ones that actually are mycelium look weak

Looks like mold to me. I had some like that looked like that. I tossed em.

It looks more like "dust" (mold) than mycelium to me. If that is mycelium, which it very well could be, it is SUPER weak. If this is occuring across multiple variations and dishes I would examine your agar recipe. Experiment with a different recipes since the mycelium may not have enough nutrients in the agar for strong growth.

Thanks for the replies!

On this plate I used 20g/L LME .... even if the LME hydrated, I feel like that should be enough nuts to have decent growth.

I first thought that I have mold going on, but after several plates, different clones, syringes, prints, etc. I feel like its suspicious that I have the same type of mold growing fucking everywhere! The other day I transferred and nothing happened at all ? super weird...

I want to switch to a different agar recipe, away from LME now. I just feel like it can't be that I have that large amount of (the same) mold from all different sources... but that's just a feeling... maybe it is possible!

LME is good for agar but I would add to your recipe. Per 1000mL of water, add.
20g Agar Agar
20g LME
2g Nutritional Yeast

Also, what environmental conditions are you keeping these dishes in?

This was my thought as well.  Either mold or really weak mycelium.  It seems like you can see the whiter mycelium right around the edges of the wedge and it just stops.  When it has a clear barrier of non growth that’s a good indication of contam.

These are with the Maltoferm LME only, no yeast, 16g per 1000mL:

I keep my plates in a spare room, 78-80F, not too much light.

Another thing I thought yesterday is, that I am always really fast with making transfers. As soon as something white(ish) shows up, I grab it... maybe I should let things grow out a little more to make a better distinctions.

I will try a little yeast the next time and also give the potato agar a shot.

and I am going to toss those btw... have to get shit done right!!

Sometimes mycelium can look really ugly on agar plates until it grows more or it gets a transfer. As long as you’re not opening them up and playing with the plates I say let the plate grow out a lot and see or do a transfer and don’t cross contaminate anything else you have going on. I’ve had really rhyzo plates come after transfers of crappy looking myc so don’t give up hope.

It’s also could just be mold and a waste of time but try to keep your spirits high haha

I usually keep a few transferred agar plates around a few days so I can see further growth or any contamination I could have brought into the plate after I opened it. Let’s me kinda see what I should expect on the new plates

I checked on the MS plates and decided, if it actually is mycelium its too weak anyways. But I normally keep my plates so maybe I can make out in the future if its mold or not.... lol

So what do you use to inoculate then?

Grow your mycelium into sand dollar size pucks and then chop them into quarters or halves to inoculate jars.

Try switching to a different agar-agar brand.
I experienced something similar to what you're describing years ago, even made a thread seeking help.
Turned out it was the agar itself. Slow and weak growth and all the cultures looked horrible, but I knew they weren't contaminated because they perked the fuck up on grains and fruited out fine aftewards.
Try it and report back.

Thanks man I will try that! I am getting fruits too and around 100% BE so I am pretty sure I don't have contamination either. Most tubs make it to the fourth flush....
I already ordered every thing new and different. Going to switch to bottled water too.
Definitely going to post if I find something out....
