Spore print to agar .

So I just received a Rustywhyte print and 20 petri dishes. Should I hydrate the spores or just put them straight to the agar. Also I  was thinking about making agar mix and then pouring them  all then using a flamed exacto knife to put the spores on each. I am waiting for some syringes as well that I want to put on these plates but I think opening the bag of plates would be a mistake before all the spores get here. I really want to do this right. Here is a picture of the petris.

Do you have an inoculation loop? I would use that to put the spores to agar. If you don't they are easily made :smile: You usually have better luck spores -> agar than syringe -> agar. Although many people have had success with syringes, me personally they always contam pretty bad.

I am going to use agar, food coloring,  honey and potato flakes for the agar mix.

Thanks I just was reading this.  Great stuff, I am going to make one now. Will copper wire work? I have a lot of thin copper wire.

I think copper should work but don't quote me. Personally I bought one off amazon for like 10$ came with two loop holders and like 50 extra disposable pre-made loops. Great investment! And also you should look into MEA. (Light malt extract agar) I tried potato agar for my first go and the viability is mediocre at best. MEA always comes out super clear for me and since I use pastyplates I need all the clear I can get haha.

If you’re relatively new, you shouldn’t try making a syringe with your spores first.

Totally do it, for practice, but to give yourself the best chance at what you’re trying to accomplish, it’s simpler to put the spores directly to agar.

You could buy a pack of cheap sterile swabs at the pharmacy and use one to scrape a bit of spores onto your agar.  I’m sure you could even just pour some out onto your agar although risk for contamination may be slightly higher with that method - the reasoning being you’re hovering your hand and a tin foil packet over your open media and shaking - contains canfall off and settle in the plate.

Using a tiny swab with concise deliberate  movements leaves less of a window for that.

Just do a search with ‘spore to agar’ and you can find people giving step by steps with pics.

You could use a copper wire. me personally I actually just use a spore syringe that's empty and heat up the needle to scrape the print. Or scalpel but I prefer the needle because it's easier to burn and sanitize. Plus It has a nice handle for grip.just make sure you wipe it down before you put it into your sab. Inoculation loops can be made from anything basic wire I have made them out of paper clips also for warning when it comes to spores a little goes a long way. I personally do not make syringes to agar. Now if I get my hands on a spore syringe I will put it in agar but it is hard to just barely get a drop on there. Hope this helps.

I find i have better control with a sterilized swab. I just PC them in a mason jar.

If you want a quick and easy inoculation loop, just use a twist tie attached to a pen. Strip the plastic coating off and twist making a loop and the end. Its that easy.

Also you do have a pressure cooker correct? I use that to sterilize everything from tools to jars (of course), But one of my favorite uses is making pastyplates. They are glad minirounds modified to use agar in. You can pick those up at the store and play around with them till you are ready to do your pours in petris :smile: I personally haven't even moved on to petris because pastyplates are fool-proof and I haven't found a need to. Eventually, I will once I have 50 transfers I need to make at once haha.

No pours are cool, but you may finally decide to switch to petris once you want to start showing off :wink:

I make pasty plates. I have 10 growing right now. I am gonna try the petris. I am doing spore prints and tissue transfer.

Ahh very nice yeah I do want to show off eventually ! I have some pretty transfers but they aren't quite picture perfect yet. And seand make sure you get one of those 500ml media bottles for pouring. Bod recommended that and it's fucking awesome. I just make up like 1000ml between two bottles chuck it in the PC and it's good for a long time :smile: I just happen to use it for no pours because I don't feel like making up agar everytime I want to make plates.

When I first did agar 4 years ago I started with pasty plates too.  Handy for sure.  I may incorporate no pours again.  I still can’t walk down that aisle in the store and not look at ziploc twist-locs and glad mini-rounds 🤣🤣. So weird.


I will say that d3’s perfect agar transfer tek (using the punch) is an instant level up.  If you go petris, you won’t regret doing it his way, especially if you’re in it to get the eye-candy like me 😳

I've been meaning to get a leather hole punch so I can try D3's method as well! Was just looking at that last night lol. I think i'm gonna buy one today. I just went and inoculated 10 BRF jars, gonna see what happens. The syringe was questionable so starting it on those guys to maybe get some good prints for grains later.

Also did some transfers and I swear the whole time I was like fuck I need that hole punch..

And yeah that bottle is the same one I have, I love it.

I would make 400 ml and just save the rest for when you need it in the fridge :smile: Never hurts to have extra. But if you want exactly I would just make 250ml, half the amount of 500ml recipe that way if you accidentally overpour on some you will still have enough for all 20 plates.
