Was hoping for some advice. First time using agar. I transferred an in vitro brf pin to agar in the 9th. I’m trying to decide where to take some samples to transfer.
Any suggestions; all the same plate

Thanks
Is Panaeolus bisporus from Washington able to grow indoors at around 72-76 degrees using modified pf tek. Also can I noc it up straight from a ms syringe. I see some grow teks on here but temp isn’t really mentioned. I’ve read the Washington strain can grow in milder climates.

I don't grow pans, but this thread is dedicated to panaeolus mushrooms and it's super active. You might try your question there if no one chimes in here. Its also worth reading through it, there is a ton of good info there.

Been seeing this a lot lately. I have done a lot of reading & trouble shooting of my SAB process already and now I’m wondering if it’s our water and that I am spraying contams into the box along with the soapy water. Lately we’ve been seeing some black crud build up on our faucet ends and in the toilets. I cleaned up a bunch yesterday in the bathroom and this morning I saw my latest pair of transfers starting to launch satellites, on day 2. That session, I felt pretty good about my process in the box and the changes I made to it. I remember a while back, some members had traced issues with their grows to their water, but that was affecting their sub. Anyway, I will try a dry box & distilled water next I guess, but anyone have any experience with this?
Also, any favorite SAB videos? It can’t hurt to keep trying to dial in my technique.
its probably something small like your scalpel handle not sterile, or you're too rough opening them. As far as videos, the best ones to watch are you own. Get a different perspective. You know what needs to be done for success.

It’s strange because this is new, I use an SAB and before I would just ISO wash my hands and be very careful and didn’t have any problems for a long time but I started to use iso washed gloves after this happened but even with a clean SAB and iso washed gloves this happens the only other thing I can think to blame is the parafilm since it’s all around the edges that’s the only thing I can make sense of. What do you guys think? Any help would be appreciated.
either your parafilm is old and cracked or you should have sanitized the plate befoee inoculation

Personally, i don't even know how many times I've gotten plastic on my tools, but its a lot. And my problems are always elsewhere.

Sorry if someone replied, I did go back through the thread but couldn't see one.
I've just made my first agar -> agar transfer today and am leaving for two weeks this afternoon. Is there any reason not to put the plates in the fridge while I'm away?

So long as the temperature is above freezing, MOST species will survive... if you can get the temperature closer to 40, theyll fair better and wont take as long to recover...
