Cleaning up a disgustingly filthy (wild) spore print?
    #26881736 - 08/15/20 02:00 PM (3 years, 5 months ago)

An old office mate sent me a spore print.  It was collected from some ox shit, in a pasture in Thailand.  If that wasn't horrible enough, the print was taken on the 1000 Bhat note.  I can't even imagine what horrors lurk on any paper currency!!

I tried the usual method, germinating them on agar.  I streaked several plates, and the things are full of mold in about 2 days.  Overtaken by mold, before any chance of the cubes germinating.  I did this with 3 different agar preps. It's not contaminating in the un-opened plates.  From what I read, trying to clean the print is an impossibility. Anything that kills mold will kill the cubes.

Are their any tricks of the trade?  I've done wild spores before, and normally I could find little clean(ish) fragments of agar.  Not these!

One option, from what I read, is to place the spores, let the mold germinate, then cover it in hot agar.  Another I was thinking about was making a syringe, and trying to get something clean in a PF cake.  Or inoculating spores or spore syringe diretly to grain and hoping I can find a clean-ish grain to extract?

Or just keep trying spores to agar? Maybe try to get the tiniest possible collection of spores and go with that?

Use antibiotic agar.  Take transfers from leading edge. Repeat as many times as necessary.

This isn't a bacteria problem I don't think.  It seems to be mold, which I don't think antibiotics will help.

Nice name though :-)  Check out my avatar
