carbon agar - contam brah

Playing around with carbon agar. Not sure if it like it or not. Can't see contams as well.

Anyway, streaked some plates with B+, KSSS, Mex/Dutch King and a PE LC. Mex/dutch was a total dud so far.

Are these prints totally garbage or what? They look terrible to me. Thoughts???

Contams are showing up fine on plates 2 and 3. Plate one might be okay, make a transfer and see how it recovers.

Thanks Pastywhyte! I see the contams on 2 and 3, just more used to the less opaque agar i guess (first time with carbon).

I'll let them grow out several more days and update :smile:

Any suggestions on to where to sector out 2 and 3? Myc is very wispy and I have a hard time determining where to transfer from. Its day 6 now (too early?). They look basically the same except about 10% larger. The B+ T1 (number 3) has a more distinct border between the 2 now. Any help is very appreciated.

There is no saving plates 2 and 3 unfortunately. They are all contam.

Plate one looks ok though. Maybe give it another day or two

Do you guys think all the spores from these prints are gonna come out this contaminated? (Logic tells me yes).

I have received a bunch i would love to work with from the same supplier. Being up north limits my print selection and I unfortunately I didn't use a trusted sponsor (but a supplier of spores since 2011).

Maybe a better question: should i continue to try agar work with these to clean them up(plates 2 and 3)?

id suggest just trading with ppl here if u want prints. that really is your best bet. i just sent some pan prints to canada and im waiting to see if they make it to the destination. it seems like everytime i send there or the uk anymore shit never arrives. not that i care about the 1.20$ i just feel bad for the ppl not getting their prints i said id give them.

i have a cpl xtra golden teacher prints that il try to send u if u wanna pm me an address but i cant promise it will make it. seems like u guys are super strict with ur mail or somethin i dont get it?? i see lots of others have no problem sending but i always do🤷🏼🤷🏼

A sample size of one plate per print isn't enough to determine that the prints are unusable. I mean I had one single spore syringe grow a couple plates of yellow boogers, then new plates that grew nothing at all, and then more plates that germinated fine. Was the syringe dirty? Probably. Was it unusable? Not at all. Did I go on to make my own prints and use those instead? Hell yes.

So too small sample size to tell i guess? I will try a few more from these and other prints and see what happens.

jcm4620 i really appreciate your generosity and i may take you up on that offer. I would love to trade but I only have GT prints that everyone already has. I have about 14 sub-species prints from the same supplier and i have only tested a few.

What i do have is an unusually aggressive PE LC (this is plate #1). Just started with this but wow the myc is fast colonizing and highly rhizo. Jars 100% in 7 days (shake at 30%) 5lb bags in about 10-12 days. The myc in the grain was the thickest, most vigorous myc i have ever seen (will take pics next grain run).

I made other LCs from the master and in 3 days each was the size of a large cotton ball (maybe 2.5-3" diameter) and had to stall them all at day 7 because they were actually growing too quickly for my timing. I will make prints (best way to make PE prints?).

You can also trade cult supplies. I have like unlimited parafilm from work, bunch of blades, SFD's, etc. So I always trade that shit for prints if I'm starting a spore collection again after tossing everything for a move.

You could try antibacterial agar for bacterial prints, but unfortunately your problem is mold, which is a bit harder to deal with.

The best you can do is make a bunch of plates and maybe try different areas of the print and hope you can get clean myc before the contam sporulates.  You can still attempt a save if it has sporulated, tho. You just need to follow up with a couple rapid transfers before the contam spores germinate.

Thanks for the tips One of Us. I am in the process of transferring now in an attempt to salvage them.  Crossing my fingers...

BTW, I have heard of a tek using a peroxide q-tip (yes we know how much everyone here loves H2O2 lol) to help mitigate mold contam during the print to agar process. Anyone have success with this?

PEROXIDE HAS NO PLACE IN THIS HOBBY

start new plates and use proper sterile technique. if u dont have sterile swabs then sterilize some the proper way with a pc. or you can do what i do and buy a box of 200 for 10$ they come in sterile packs of 2. your problem is most likely coming from contammed print or operator error within your sterile procedure.

Also, if I have a really dirty print, I sometimes use a more involved streaking pattern, such as the one detailed in c10's agar guide. It seems to spread out the print more across the plate and gives a better chance of getting a clean section for transfer.

ya absolutely👍👍 il go his route also or sometimes il just zigzag over a cpl plates like i did with these pan aussie tropicalis/rdu

these were actually the best looking and fastes streak plates ive ever had. these pics were taken at the 36 hour mark. and i had already taken transfers just after seeing germination just after the 24 hr mark i took 10 transfers from each plate and you cant hardly even tell



then here are the same plates at 48hrs from streaking. i am still amazed at how fast they were and how clean and good the growth looked.
