Ecuadorian Cubensis Shoe Boxes

Variety: Ecuadorian Cubensis

Inoculation Process: MS syringe > MEA > Master Grain jar > Grain2Grain

Grain Used: Whole Grain (Producer's Pride) Boiled Via BOD's TEK

Substrate: 100% Coir prepared Via BOD's TEK

Grow Method: Shoeboxes Via Shaper's SFF TEK




Inoculation Prep

6/22/20: I introduced Multi-Spore Solution to Malt Extract Agar and allowed germination.


6/29/20: Transferred to clean MEA plates and allowed solid growth.


7/7/20: One plate was lost due to contamination (not pictured).


7/8/20: AGAR added to grain jars.




Grain Jar Prep

Producer's Pride Whole Oats horse feed.

I washed the oats a few times to remove the majority of the stems, hay, and random kernels that are inevitable in the bag. Boiled for 25-30 minutes on a steady boil. Pressure cooked for 2 hours at 15 PSI. The jars are 1 Quart Mason Jars with a 1/4 inch hole punched in the lid and stuffed with poly fill. I filled the jars 3/4 full with the boiled grain. These will be master jars to use for Grain2Grain inoculation for 9 more jars. I did the Tic-Tac-Tiger Drop method to inoculate the jars, cutting a Tic-Tac-Toe board into the AGAR plate to allow for more inoculation points. This is my first attempt to inoculate with AGAR. All work was done inside of a SAB.


NEXT STOP! COLONIZATION STATION!


7/14/20: First shake on Master Jars (+/-25% colonization)

7/17/20: Second shake on Master Jars  (+/-75% colonization)
              Tossed one jar due to contamination. (Greenish/Yellow growth)

              A fellow mycologist (LadysKnight) called contamination on the
              middle agar plate, I disagreed with him. Unfortunately I did
              not think far enough ahead to mark which jar came from which
              plate, but I believe it is safe to assume LadysKnight was
              correct in his assertation. And the plate which was
              contaminated led to the lost jar.

7/21/20: Shake and Bake (Grain to Grain transfers) 2>8


              P.S.  The grain inoculated with agar colonized one week faster
                      than my last group of jars which were inoculated with
                      multi-spore syringe. I'm excited to see the
                      colonization time for the Grain to Grain inoculation
                      process.

7/24/20: Grain to Grain jars have recovered nicely.


7/26/20: First shake on Grain to Grain (+/- 50% colonization)

SPAWN IT UP BITCHES!


7/29/20: 9 days from Grain to Grain inoculation to 100% colonization.
              Two jars reached 100% colonization and were spawned to pure
              coir substrate. A ratio of 1:1 was used. One jar was lost to
              green mold, my guess is from bad technique during Grain to
              Grain inoculation.

8/4/20:  4 jars at 100% colonization spawned today to 1:1 ration pure
              coir with 1/4 inch top layer.

8/5/20:  Updated pictures of colonization from the two spawned jars
              spawned on 7/29/20.

8/18/20:  These Ecuadorian boxes are moving tragically slow. The boxes
                that were spawned 7/29/20 have started pinning, very few pins
                though.



                The four boxes spawned on 8/4/20 have knots showing up
                and look a bit healthier than the first two.

Looks awesome! I'll be following this for my fist shoe box in a week or so. Using Popcorn with coir/verm in a 6 qt shoe box. Popcorn is looking good, time to shake soon. Going G2G with two of the faster jars. Looks great. Are you using just a flipped lid? Did you let it colonize before adding much FAE or just go straight to full FAE as used when fruiting? I'm getting that shoe boxes are sort of "always fruiting conditions" due to the design, but I have seen people put the lid on tight and try to limit some FAE at first to let it colonize a few extra days.

I follow Shaper's tek for the most part. Shaper's Shoe Boxes.

I have a different style of shoe box so the lid does not have the clips. The grey lid boxes from Walmart. I clip them on and leave them until pins are fully formed and growing. Then I pop the lid loose and simply set it on top. I've found that if I leave the lids clipped in place on this particular style shoe box, my pins start getting fuzzy feet once they are about 2 inches. And that's that, I leave the lid setting on top but not clicked until harvest. Once the shrooms grow to the lid they push it up to get as much room as they need to grow.

This was my last set of shoe boxes grown to completion. You can see how that one rolled along as well. Soupy's Cambos.

It's not nearly as indepth with the description of what I did as this one is. But I did post pictures of it through colonization and fruiting. The spawn starts on page 3 if you want to skip all the grain jar pictures.

Can you explain  where the contamination was visible on the middle plate.

How long for full colonization of your agar to grain jars. 4.5 weeks?

The jars inoculated with agar took 13 days to reach full colonization.

I personally didn't think the middle agar dish was contaminated, I thought it was all good mycelium. The shroomerite LadysKnight mentioned that it was contaminated. I still don't know for sure as I didn't mark which agar went into which jar, I just assume he was correct due to losing one jar to contamination. The contaminate that showed up in my jar was green mold. This is the post where he brought it up.

I have an extremely limited experience in this hobby as I have only been working with agar long enough to make two transfers, but my educated opinion would be to let them grow out more. The amount of mycelium on they plate would just take a while to colonize. Even just 3 more days on the agar and it will grow out a good bit more giving a significant difference in your jars colonization time.

Do you mean those agar transfers pictured in this grow? They were all spawned to grain in the growlog. Now I am in the process of making agar plates with a spore print I took from a Cambodian on my last grow. There is nothing to show for that yet.

What I did with the agar pictured. Firstly I made a SAB (Still Air Box), a big clear monotub with arm holes cut in it. I shook a multi-spore syringe until the main clump of spores was busted up and shook it for a bit more. Then, without attaching the needle that comes with the syringe I put one drop on each agar plate and spread in across the agar in different methods. From letting it roll across the agar just by tilting it, making a straight line down using a homemade inoculation loop, and doing zigzags using the inoculation loop. All of these methods germinated but I much preferred the single line down the plate when I came time to transfer. That,s to only time I've inoculated agar with MS syringe so it would be wise to look up some others who have more experience with it. Bod is the one I studied before hand.

First lesson I learned about the sab is after you spray it and set it up, make sure all air sources in the room are turned off, A/C, fan, windows, and doors. Then let the box sit for 10 minutes or so before doing work inside, so the air inside the box can settle allowing most contaminates to stick the soapy water.

My most appreciated one liner in the hobby is, "Mycology takes patience". In ever thing I do with mycology, now, I do it slowly and deliberately. I've finally made it through the helicopter phase, hovering over my grows, luckily. And it is now more enjoyable since I'm not steadily stressing on it. When it does come time to do work, it's been long enough since I touched my goodies and I am excited to sit down, take my time, and enjoy every moment of what I have to do.

I believe the contam is on that second plate between 10 & 12 o'clock. Where the myc is raised up higher than the rest. But I definitely do not as as much experience as a lot of these great folks here.
