how much it affects and what changes in different thickness of the growth substrate on a monotube?

What are the correct proportions?

And in practice What changes with a low substrate example 2 cm has a substrate high example 10 cm?

The typical ratio is 2:1 sub to spawn, mine end up around 100mm thick with a 6-8mm casing in my 56qt tubs. I'd recommend following a standard TEK and ratios.

Thin substrate = more surface to volume = dry out quick

Up to 4" for monos, at least 2" for shoeboxes.
