Decided today that I needed to take my transfers out of the wild culture dish. I noticed the mold colony has grown much more in the last 24 hours and if I wait another day I’m afraid it will be too late. Plus I think this will be fun to see if I can isolate out a few clean cultures from a dirty plate and if not I haven’t lost much. Here is the donor, this was my first batch of agar and I messed up and splashed the liquid agar on the lids of a few of them and messed them up. You can see the green mold at the top of the dish.
 I carefully opened up the dish and took small sections out of what to me seemed the strongest, it also made me feel better that it wasn’t the side directly oriented towards the mold. I spritzed my hands and scalpel with alcohol in between every transfer along with flame sterilization. Fingers crossed

Looking for opinions on this growth. Would you keep moving forward with this particular growth? It seems to be very light although it does seem well organized. I’m sure someone can weigh in with some experience

Also, an update on the wild culture transfers. Either I waited too long or I need to improve on my sterile tech some or both. All three transfers had mold colonies pop up all over the plates and I tossed them so that venture is over. Oh well, I didn’t have high hopes for them to begin with

Is also like opinions on these transfers. Maybe I’m paranoid but does this white whispy growth look normal or moldy? It seems to be consistent with this set of plates, hoping I didn’t wait too long to transfer them out. Am I paranoid or do they look okay? The third one with the satellite has me suspect..

I guess I’ll go with my gut lol. Anyway making another set of 20 plates today, I have some t2s to make and maybe some t3s, I need to go in there after I pour and see which one’s i want to transfer yet. Anyone else use a coffee pot for a water bath? It’s not well insulated but pretty quickly my coffee maker will make a half pot of 150degree water, fill, pour and fill with cool water and it’s at about 120 degrees. It works well for bringing the media bottle down to a close to working temp pretty quick and holding it there for a small amount of time

Dropped a QT of grain into 2 QTs of coir last night. I’m really excited, this is the first time I’ve been able to spawn grain to substrate. Also I have a hand full of jars that will be ready for substrate in the coming week and 2 more that are nearly ready for a shake

Shoebox seems to be under way. 2 days after spawn, seeing the mycelium bounce back nicely

First shoebox spawned on 8/8 with 1:2 spawn ratio in coir with just enough coir to cover the grains. The second shoebox was spawned on 8/14, same ratio of coir without coir covering the grain. I have some Jars of Jedi’s that will be ready to spawn this week, and I plan to do the same with them as well

Wanted to share some agar cultures I’ve been working with. I started out with around 20 germ plates and I have culled hard to narrow it down to these few that will go to grain in a day or two. I’ve already taking some transfers from the GT as I plan to do with the others to store away in case they turn out to be good cultures in substrate. Sorry for the condensation plates. I’m still working on that

Shoebox of GT 18 days after spawn
 This is MSS>Popcorn>Coir. A 1:2 ratio with about 1/2 a Qt of coir on top to cover the grains. I’m struggling on this one to keep the moisture right without flooding the surface. I need a new spray bottle at the moment
