What are these little dots on the mycelium (agar)? Are these knots?

Hey guys.
Are there knots? I've never seen this on my agar plates before.
I was going to inocculate them today to grain in about 10 hours. Are these okay to throw in?

they look like knots to me. i usually donโ€™t get knotting on my plates unless they sit for a really long time tho. usually the entire plate is covered before it happens

Holly fuck I hope they are not knots already. It's really weird because I made that transfer just one week ago.
Will hurry up today to inocculate it asap!:eek:

has it colonized the plate evenly?? it looks strange in the back of that last pic to me but it may just be the angle or something

if it does end up being a clean plate... iโ€™m pretty sure you can use it to inoculate. iโ€™ve seen people who have said theyโ€™ve spawned jars that had pins & it went fine

you could also wait for the mushrooms to form & then take a pin as a clone to another agar plate ...

Knots

Is there a difference in the % nutrients from the transferred block and the current plate?

I often see knots developing on the outer perimeter when the leading mycelium touch the edges. By leading I mean the faint whisper of glass like fibers scouting out ahead of the hard edge.

Iโ€™ll see knots closer to the transfer block if:
    There is a drop in nutrients or moist compared to the block
      The nutrients in the plate are less appealing compared to the block

Knotting plates are fine just cut them in sections.

The real question was asked above. Is there less growth under the words of the plate? If so why?

The knots close the the block could also be competitive behavior.

I've already inoculated it.
And yes, as you could see, it wasn't colonized evenly. In a sector (third picture) there was a notorious delay with tormentose behavior, which was the only sector that did not inoculate just in case.
I think it could be just a genetic matter or something like that (the plate is multispore, it's a first transfer from a print).
And yeah, you are right Spaghetti, I think those little knots could be from the change of media! The print was inoculated on dog food agar, let it grow two weeks and then transfered to regular LME. Maybe that could be the cause!:rolleyes:

it could have been contamination which is why i asked abt it. iโ€™m not sure that one transfer from multispore is necessarily enough to ensure a clean culture unless you get really lucky or know what youโ€™re doing & what to look for. that tomentose growth coulda just been a genetic thing or it coulda been the mycelium growing around/over a contam. my plates that have knotted/pinned were way overgrown when it happened. if you cut around it you MAY be ok, but contams can be pretty sneaky.

Those look like hyphal knots to me. Looks good btw! ๐Ÿ‘

I would still use that as inoculant, to be honest though I have never made spawn with agar yet. Still learning. It doesn't have any mushroom bodies that could rot though.

My plates right now have the same little knots forming after just over a week.

I'm pretty sure it's normal :thumbup:
