Issues with starting with spores prints

Hi everyone. For people who don't know me, I want to warn you that I live in Central America and I can't easily receive parcels by mail.
For this reason I have to work with spores, because they are easy to send and receive.
But I have many problems with spores. I can't grow them even though i've seen the Bods video 1000 times. I made 40 gerbers and only 3 have been successful but I'm not sure why I have to bear them again.
I use gerber containers instead of petri dishes because they are reusable but they are deep and difficult to open.

1) I don't like to open the gerber container 2 times (once to cool the inoculation loop, and once to put the spores). Maybe it's better to keep another gerber container with agar agar always open to cool the inoculation loop, and then open the other container only once to put the spores on it.

2) I put the spores in the center of the container because at least I am sure that if the mycelium is born from the center it is the right one, while if it comes out from the edges it could be contamination. But I see the process from spores to mycelium very very slow. How many days does it take to grow the mycelium from the spores?

3) can you help me with generals tips and suggestions?

it can take a week or more for spores to germinate on plates. you’ll have to be patient, as working with spores is generally just pretty slow. sometimes i see growth in two days, other times it takes a week or so. also, i think your agar composition can affect how readily spores germinate. i don’t know a ton about that but have seen it mentioned. when you say you made 40 plates & only 3 worked, do you mean nothing grew on the other 37? or are you running into contamination?

my suggestion for agar would be to flip the jars or whatever upside down so your lid is on the bottom of the SAB. make sure the lid is off, so you can pick the jar up & flip it over when you’re gonna put spores down or make a transfer. does that make sense? that way the jar isn’t just open to the air the entire time you need it out. the spore swabbing zif linked you is good. i usually just use a paper clip i bent into a loop, put it in my scalpel holder thing & fire it up. you shouldn’t really have an issue double dipping the spore loop in the same jar to cool it. i do it all the time & never have an issue. it’s always good to take precaution with contamination, but i don’t think this specifically is what’s giving you issues

general tips? just be patient & willing to learn. you’ll get exponentially better with time & experience. most people fuckin suck their first few grows, myself definitely included. once you get the hang of it it’ll start to become second nature. also don’t hover your hands over open jars or insides of lids that are sitting on the bottom of your SAB. you don’t want contaminates falling from your hands

You say you can't grow on agar. Just what is the problem? Contamination? No growth? You can use glass petri dishes, they're reusable. You could make your own spore syringe from the print and directly inoculate to grain. Far easier than petri dishes.

37 lost for contamination or nothing growing or something growing but not where i put the spores.
very good idea to flip the container .

difficult to find glass petri dish where i live Gerbers are fine. i just have to practise more and more. Thanks.

It uses Glad Mini Rounds instead of Petri dishes. You just need to cut a hole and use any type of filter material. They are very easy to use, I doubt I'll ever stop using these in favor of Petri dishes

What agar recipe are you using? And are you using a pressure cooker to sterilize your agar jars?

100 ml filtered water 
2,25 g agar agar
1,75 g malt extract
this is for 6/7 gerber
i know that post. i red many times. Gerbers containers are very similar but are done with glass. And i can find them in any shops because is Neste baby food.

That's a pretty stiff agar mix. Maybe try 1.5 to 1.75. I have a very hard time germinating dry spores on stiff agar.

this recipe works well for agar agar transfer. But i was wondering too if is to stiff for spores.
i can try to put less agar agar.

Why not make a syringe with the spores and do a pf tek?

Working with spores on agar can be tricky. Many have success with it, but lots of others refuse to go spores to agar for multiple reasons.

I think you'd be much better off sending those spores into some brf cakes, fruit those or spawn to bulk, then clone the mushrooms onto your agar. If you do it this way, you'll also have much more trustworthy mycelium to inoculate grain with. When going from spore to agar, you really won't know that the myc your isolating will even produce much for you.

yes , is a good idea... but i am not doing because i never did it. I will try in the future, but right now i feel more confortable with agar agar.
from the agar agar to see fruit is 1 1/2 months. I have to wait to see this time to see what kind of fruit i have got
