First Attempt at Cultivating Mushrooms - Mini Mono

When my country entered lockdown due to COVID-19, I realized I would have ample amounts of free time to explore things I normally would not have time for.  This was a great opportunity to delve into the world of mycology- a hobby I've admired from afar for a while.

After religiously reading teks and forums on this website for the better part of two weeks, I decided on a plan of action.  First, I would isolate clean cultures from an MS syringe on to agar, then I would inoculate grain spawn, which I would then spawn to a monotub.  All sterile procedures were performed in a SAB.

The SAB I created deviated from the standard.  I cut oblong holes, measuring 5.5 inches wide and 10 inches in height.  This hole shape has allowed for extra comfort when operating inside of the SAB.


My work set up.


The agar recipe I used came from Frank's Media Journal.  10 g of agar agar, 10 g of LME, and 1 g of yeast flakes per 500 mL.  I added food colouring to the media, pulling inspiration from c10's agar guide.  I PC'd the agar for 30 mins at 15-18 PSI.  Note** I used 500 mL Pyrex Media Bottles as the container for the agar.  Well worth the investment.  One of my favourite pieces of equipment

After dropping Golden Teacher spore solution into a shot glass, I used the "z" streak method to transfer the spore to agar as suggested by bodhisatta.  As expected, the spores were not perfectly clean, and transfers were required within a week.


After 3 rounds of transfers, I had cultures that were exhibiting decent rhizomorphic growth.  At this point, I had about 40 plates that looked like this.  Way more than I needed.


Once I had clean cultures, I purchased whole oats.  I followed eat's oat prep, as I was drawn to the prospect of foregoing the soaking process.  After letting the oats sit in boiled water for 45 mins, and then PC'ing for 90 mins, my oats came out perfect.

Due to my surplus of agar plates, I dedicated nearly a whole plate to each grain jar when it came time to inoculate.  Part of my rationale behind this was that I was working from an MS syringe, and wanted to include a fair amount of genetic diversity.  I didn't want to run the risk of using a few number of cultures that might turn out to be poor fruiters.

During the inoculation process, I performed the tic tac toe method of cutting agar with a scalpel.  I then would stab 3-4 pieces at a time and drop them into the grain jar.  Albeit, some pieces stuck to the sides of the grain jars, but I did my best to distribute them throughout.  The goal was to maximize the number of inoculation points.  At 20-30% colonization, I gave the jars a shake, and they rebounded with vigor.

Once my grain spawn was 100% colonized, I spawned 4 quarts of spawn to 6 quarts of coir in a 28qt Sterilite tub.  I elected to go the "unmodded" route even though the model of the tub was not the exact same as the one bod suggests. 

This is where I ran into my first real issue.  Life picked up again, and during the first week of colonization, I was unable to tend to my tub frequently.  I had immediately placed the tub into "fruiting conditions", with the lid flipped upside down.  This led to portions of the surface drying out.

The result of this was decent pinning on top...and a shit load of pinning on the North, South, East, and West sides of the tub haha.



I do believe that these side pins were a direct result of poor surface conditions.  I currently have a new tub on the go, that I "pseudocased" with approximately a 1/8 inch of coir, and waited to fruit until the tub had fully colonized.  That tub's surface conditions have remained prime throughout, and pins are predominantly popping on the surface.


Nonetheless, healthy-looking fruits have been produced, and I'm super proud of my first effort in this fascinating hobby.  Thank you to the many contributors on this site that provide high-level education to make this hobby accessible for all.

I'm certain that the sides receiving light wasn't the issue.  I have a second tub on the go right now that does not have a liner and this time I've kept up with it's surface conditions.  Low and behold my side pinning issue has been rectified. Surface conditions must be optimal to avoid side pinning.
