Opinion on clear growth agar

Noticing clear growth developing on agar after 4 days of transfer

1.Does this clear growth look like contamination?

2. is this a tomentose sector?

3. is this new growth that has not turned white?

4. Am i safe to go to grain or should i wait.

I have tried several different agar recipes and nutrient strengths, this is the fourth transfer from spores. After i retired my old strain after seeing similar growth can anyone help me ? has anyone seen this before ?

I don't know about other users but I tend to see this from time to time. Usually is the print (or my technique) wasn't the cleanest. Just transfer from the healthy looking growth to the next plate.

I have had healthy growth on the second transfer and other times end up doing 7+ transfers. I would say that the look of a MS syringe on agar made probably the most bizarre looking growth like that in my experience.

Thanks for the response. More transfers would usually be my go to solution. After runnning successfully for years with another culture i moved several months ago and brought my clean plates. After setting up in my new spot i removed the plates from the fridge and did a transfer as i usuallly do(was at transfer 12 on the original gt culture). This produced similar growth as to what i have posted pictures of. After doing another 4 transfers on that culture all with different agar recipies, types, brands etc i decided to trash that and start again from spore syringe. These pictures are cambo i recently started in the same new place and as i said have hit transfer 4 and is showing the same weird growth. If your suggesting more transfers do you have any insight into what im transferring away from.

It's hard to tell from the pics.

Have you tried rye soak water agar? I was able to get rid of a similar growth that I believe was mold by transferring to rye soak agar, then it stayed gone after transferring back to malt extract.

The mold is the lighter ring behind the splotchy looking mycelium. You can see the mycelium trying to out run the mold, there are several sectors between 2 and 3 o'clock in the last pic that just may make it.

:whathesaid: try to get a piece that has outrun the mold and keep transferring until you got away from it completely.

  If you get a fast enough piece it might outrun the mold.

The picture quality isn't great but, to me it looks like mycelium that could be growing into the agar. The transparent sectors have the typical rings you see on mycelium and the lines all seem to grow out in line with the whiter sectors. When in doubt grow it out. Monitoring it is the best way to learn.
