Avery and Nepal plate transfers
    #26845629 - 07/26/20 07:35 AM (3 years, 6 months ago)

Hey guys I am doing my first agar cultures and I need some help figuring out if this is mold or mycelium.

I inoculated on the 19th July and 3 days later I could see germination. I did read somewhere that spore germination happens between 4 and 30 days. 3 days is too early? Where I am right now it's summer and it's about 35 Cº +- outside. Can the cubs spores germinate earlier in warmer conditions?

I poured 19 plates (not enough agar for the 20th plate). Used SAB, sterile technique the best I could: cleaned SAB, used gloves, 70% alcohol, never go over sterilized media with hands/gloves, flame sterilized the loop etc.

I inoculated 10 plates and have other 9 which are still not inoculated (1 got contaminated with some smelly transparent bacteria I guess).

Also I dropped some blue food grade ink. It looks like the myc or mold is eating the blue. Is this normal? If so is there any type of food colorant that the myc won't eat?

Anyway appreciate your input. Pics are below.

4 plates inoculated with Cubensis Nepal variety, here's the pics:

Heres a pic I took from the side of one of the plates:

And here's 4 plates inoculated with Cubensis Avery's Albino variety. This spores came with swabs so for this I did not use the inoculation loop, instead I roiled the swab all over, also because I wasn't sure if the spores were being dropped on the media. I could not find a TEK for Swabs and Agar so I did the best I could:

Oh and here's 1 plate with Cubensis Escondido:

I got another plate with Escondido but no germination happened yet and I see no bacteria or molds etc. Could the spores be dead or something?

It all looks like myc...maybe some areas of suspect in that 3rd Avery plate, but other than that just looks like some good tomentose growth. Take some transfers.

Mush myc loves to eat food coloring.

Hey Tomato thank you for taking the time to read this and for the tip.

I did some transfers to 8 new plates, 5 Averys and 3 Nepal. Will report back once I have something going on those plates.

Hey here's pics from my transfers. I chose the best looking pieces on the first plates and transferred to new ones. I'm still a bit worried that this might be mold. The myc looks fluffy and light. It looks different from most agar plates I see being posted here where the myc is thick, white and ropey.

What do you guys think?

So here are the pics:
